Abstract

Setiap tahunnya, kebutuhan pakaian di Indonesia terus meningkat. Dari 209.300 ton pada tahun 2009 menjadi 308.400 pada tahun 2014. Peningkatan kebutuhan pakaian ini dapat menyebabkan masalah lingkungan seperti polusi warna di sungai Citarum. Pewarna tekstil yang digunakan disebut pewarna azo. Pewarna ini berbahaya karena dapat mengakibatkan kanker dan masalah kesehatan lain. Dalam penelitian ini, khamir dari ragi tapai digunakan sebagai salah satu usaha biologis untuk memecahkan masalah ini. Jenis khamir ini tidak berbahaya dan telah menunjukkan potensi dekolorisasi yang baik Pada penelitian ini, khamir diisolasi dan ditapiskan pada 13 pewarna azo untuk mengetahui warna mana yang dapat didekolorisasi paling baik. Dari hasil isolasi, khamir yang diperoleh diduga memiliki genus Pichia. Pichia diinkubasi pada suhu 30 °C selama 24 jam. Hasil dari penapisan tersebut menunjukkan pewarna congo red yang terdekolorisasi sebanyak 80% dalam waktu 24 jam. Namun, hasil pembacaan spektrofotometer masih menunjukkan adanya gugus benzena yang berbahaya bagi manusia. Studi lebih lanjut menunjukkan suhu dan pH optimum dekolorisasi adalah 35 °C dan 4. Pichia yang diisolasi dapat menghasilkan enzim azo reduktase setelah masa inkubasi tiga jam. Hasil penelitian ini menunjukkan bahwa khamir yang diisolasi dari ragi tapai diduga memiliki genus Pichia. Pichia yang diinkubasi dapat digunakan untuk mendekolorisasi pewarna azo seperti congo red dengan efektivitas dekolorisasi 80% dalam kurun waktu 24 jam. Namun, produk akhir dari dekolorisasi masih menunjukkan zat berbahaya bagi manusia. Studi lebih lanjut diperlukan untuk mengetahui spesies dari khamir yang diisolasi secara pasti, dan adanya kemungkinan mikroba lain pada ragi tapai yang berpotensi dalam mendekolorisasi pewarna azo lain dan dapat mengurangi kadar berbahaya dari pewarna azo. / Every year, the need for new clothing in Indonesia keep rising. From 209.300 tons in 2009, to 308.400 tons in 2014. This massive increase of clothing production can lead to many environment problems, including color and toxic pollution in Citarum river. The dye that is used for clothing is called azo dye and it contain toxin called benzene which can lead to cancer and other health problems. In this study, yeast from tapai is used as a biological approach to solve this problem. This type of yeast is not pathogenic and has shown good potential for azo dye decolorization. In this study, the yeast is isolated and screened through 13 different azo dye and incubated for 24 hours in 30 °C to find out which color it can decolorize the most. The yeast suspected genus is Pichia. From 13 different dye, Pichia can effectively decolorize congo red with decolorization rate of 80% in 24 hours. However, the spectrophotometer reading shows that the byproduct of decolorization still contain dangerous substances such as benzene. Further study reveals Pichia has the optimum temperature of 35 °C and pH of 4 to optimize the decolorization rate. Pichia has also shown to produce azo reductase enzyme three hours prior incubation. This study has shown that the isolated yeast from tapai is suspected to be Pichia. This Pichia can be used for decolorization of azo dye such as congo red with decolorization rate of 80% in 24 hours. However, the byproduct of decolorization still contain toxic material. Further study is required to know the exact species of the yeast and other microbes that contained in tapai yeast that may contribute to decolorization of other azo dye and reduction of toxic compound in the dye.
