Abstract

Imunomodulator merupakan agen kimia yang dapat memodulasi sistem imun dengan menstimulasi produksi antibodi atau inhibisi aktivitas sel darah putih. Pengunaan imunomodulator dapat berpotensi dalam meningkatkan pertahanan tubuh terhadap infeksi patogen penyebab penyakit. Probiotik dapat diaplikasikan pada host dengan metode oral (sonde) atau intraperitoneal injection (IP). Melalui metode sonde, probiotik yang masuk ke dalam tubuh akan melawati beberapa tahap dan memacu respon imun dari host. Probiotik yang masuk ke dalam tubuh akan berinteraksi dengan sel imun di peyer’s patch yang merupakan jaringan limfatik pada ileum usus halus. Pada metode IP, probiotik berinteraksi dengan host dengan sistem imun sistemik dalam darah dan limpa. Probiotik dalam darah dapat memodulasi aktivitas sel T melalui teichoic acid. Uji yang digunakan dalam penelitian ini adalah perhitungan ratarata sel darah putih (WBC count) dan rata-rata titer antibodi total pada hari ke- 0, ke-5, ke-10 dan ke-15 setelah penginjeksian antigen (sRBC). Pemberian L. fermentum E5 secara IP memiliki jumlah rata-rata jumlah sel darah putih yang lebih tinggi berbeda secara signifikan dibandingkan kelompok IP NaCl 0,8%, sonde L. fermentum E5, sonde NaCl 0,8%, dan tanpa perlakuan. WBC count kelompok L. fermentum memiliki peak pada hari ke-0 dan mengalami peningkatan kembali di hari ke-10. Titer antibodi total kelompok L. fermentum meningkat pada hari ke-10 (log2 titer 7,5) dan ke-15 (log2 titer 10,25). / Immunomodulator is a chemical agent that is capable of modulating the immune system to stimulate antibody production or promote anti-inflammation. Immunomodulators have the potential to prime the body’s defences prior to pathogen infection. Probiotics can be applied to its host via oral and injection to the peritoneal cavity or intraperitoneal injection (IP). When probiotics were consumed and went through the oral pathway, it goes through several phases before inducing a response from its host. Probiotics that goes through oral pathway will interact with immune cells in the payer’s patch, which functions as a lymph node at the ileum of the small intestine. Probiotics that applied with injection via peritoneal cavity will interact with immune cells that resides in the blood circulation and the lymph nodes, as the peritoneal cavity contains blood vessels and lymph nodes. From this interaction, probiotic can modulate T cells activity with teichoic acid. Parameters that were be measured in this research are white blood cell count and antibody concentration in blood serum. Data was collected on the day of antigen injection, and on the fifth, tenth and fifteenth day after antigen injection. Results showed mice that was injected with L. fermentum E5 will have a significantly increased white blood cell count, compared to other treatment and placebo group, and as well as its antibody concentration, which showed agglutination up to log2 10,25.
