Abstract

Isolasi dan karakterisasi bakteri dari lumpur aktif yang berasal dari TPST Bantar Gebang dilakukan untuk mengisolasi bakteri yang memiliki kemampuan untuk mendegradasi benzena. Kontaminasi senyawa aromatik pada air tawar dan udara menjadi salah satu isu terbesar di Indonesia. Oleh karena itu dilakukan penelitian untuk melihat potensi aktifitas biodegradasi bakteri terhadap benzena. Terdapat 3 bakteri yang telah diisolasi yang berpotensi untuk mendegradasi benzena yang ditandakan oleh pembentukan senyawa fenolik menggunakan TLC, dan peningkatan spektrum absorbansi terhadap benzena menggunakan UV-Vis spektrofotometri. Perbandingan spektrum absorbansi dari setiap sampel menggunakan isolat bakteri yang diperoleh dilakukan untuk menduga jumlah senyawa larut air yang berada pada sampel. Digunakan GCMS untuk mengidentifikasi senyawa larut air yang terbentuk saat dilakukan biodegradasi benzena seperti benzoyl-CoA, o-benzokuinon, p-benzokuinon, fenol, dan katekol. / The isolation and characterization of bacteria from active sludge retrieved from TPST Bantar Gebang were done in order to find bacteria able to biodegrade benzene. Contamination of aromatic compounds in the air and ground water is one of the biggsest issues in Indonesia. This research aims to observe the potential ability of isolated bacteria to biodegrade benzene. Three isolates that were retrieved showed potential in biodegrading benzene which was signified by the presence of phenolic compounds by TLC, and an overall increase of absorbancy spectrum towards benzene using UV-VIS spectrophotometry. The comparison of absorbancy spectrum of each bacterial isolates was done in order to predict the amount of soluble compounds produced. GC-MS was used in order to identify soluble compounds present after the biodegradation of benzene such as benzoyl-CoA, 0-benzoquinone, pbenzoquinone, phenol, and catechol.
