sistem pertahanan tubuh. Penelitian tentang manfaat dari probiotik yang dilakukan secara in vivo semakin berkembang. Namun, metode dalam pemberian probiotik melalui sonde dapat menyebabkan stress pada mencit itu sendiri dikarenakan kontak langsung secara berkala kepada mencit, sehingga penelitian suplementasi pakan mencit dengan L. fermentum E5 yang dimikroenkapsulasi dengan natrium alginat-kitosan perlu dilakukan sebagai alternatif dalam pemberian probiotik yang tidak menyebabkan stres pada mencit. Metode yang dilakukan dalam penelitian ini adalah pembuatan mikrokapsul menggunakan bahan alginat, mikroenkapsulasi probiotik, proses pengeringan, analisis viabilitas sebelum, sesudah mikroenkapsulasi, dan penyimpanan, reformulasi pakan mencit dengan penambahan L. fermentum E5 yang dimikroenkapsulasi dengan alginat-kitosan, dan uji viabilitas mikrokapsul pada pakan mencit selama penyimpanan. Pembuatan mikrokapsul pada penelitian ini menggunakan kombinasi konsentrasi alginat/CaCl2/kitosan yang optimum yaitu 4%/4%/0,75%, dan durasi pengeringan mikrokapsul yang paling optimum adalah 48 jam. Pada uji efisiensi enkapsulasi mikrokapsul L. fermentum E5 sebesar 93,96±4,09%. Pada reformulasi pakan mencit, didapatkan penggunaan konsentrasi binder yang paling baik adalah alginat dengan konsentrasi 3%, dan durasi pengeringan 180 menit. Pada uji viabilitas penyimpanan pakan yang telah direformulasi dengan L. fermentum E5 pada H+28, didapatkan survival rate sebesar ±82,4% pada suhu ruang yang masih memenuhi dosis standar yang direkomendasikan dalam pengkonsumsian probiotik / Probiotics is one of the functional foods that can improve the body's defense system. Research on the benefits of probiotics conducted in vivo is growing. However, the method of administering probiotics via oral gavage may cause stress to tested mice itself due to periodic direct contact. Hence, an alternative probiotic delivery system by using mice fodder with microencapsulated L. fermentum E5 with sodium alginate-chitosan since this way does not cause stress in mice. This research included the making of microcapsules using alginate, microencapsulation of probiotics, drying process, viability analysis before and after microencapsulation, and during storage, reformulation of mice fodder using microencapsulated L. fermentum E5, and viability analysis on microencapsulated probiotic incorporated in mice fodder during storage. The best combination of alginate / CaCl2 / chitosan concentration to prepare microcapsules was 4% / 4% / 0.75%, respectively. The optimum duration for drying was 48 hours. The encapsulation efficiency of microcapsule L. fermentum E5 was 93,96±4,09. In the reformulation of mice fodder, alginate with 3% concentration was the best binder, and the best duration of drying was 180 min. Storage viability showed that 82.4% of L. fermentum E5 in reformulated fodder survived after 28 days of at room temperature.
