Abstract

In this experiment, a high electrical voltage (high voltage category) is modulated to preserve milk. The method that uses a high voltage is PEF (Pulsed Electric Field), where in order to kill microbes, this method uses pulsed electric field. The parameters studied on the effect in killing microbes are the voltage, treatment duration, and polarity. By using an Arduino module, treatment duration and polarity can be controlled. The Arduino modules connected to low-voltage driver circuit and then using IR LED and phototransistor can control the high voltage Hbridge circuits that is galvanic isolated. The maximum voltage in the H-Bridge circuit is limited 1.3 kV due to MOSFET breakdown capacity. Therefore, the high voltage generator circuit need to be modified in order to avoid damaging the MOSFETs. Experiments were carried out by connecting 8 high voltage circuits in series which give 5698.97 Vrms with a modified sine wave with frequency of 454.5 Hz for 0,038 second of effective time (treatment duration). This experiment was carried out on a fresh milk that was milked from Bogor ranch. Milk that already being treated was analyzed using TPC (Total Plate Count) method. The degree of microbes death from TPC method using PCA (Plate Count Agar) on dilution of 10-3 between controlled samples and samples that are treated with PEF with 3.87 x 106 Vrms2second/cm2 dose is TNTC (Too Numerous To Count). On dilution 10-4, the degree of microbes death with same treatment is 54.34%. Degree of microbes death using VRBA on dilution of 10-3 between controlled sample and samples that are treated with PEF with 3.87 x 106 Vrms2second/cm2 dose is 89.81%. For 10-4 dilution, the result for degree of death with the same treatment is 61.76%. These results show that the high electric field treatment makes some bacteria die. / Dalam eksperimen ini, digunakan tegangan listrik yang tinggi (kategori high voltage) yang akan diubah-ubah untuk mengawetkan susu. Metode yang menggunakan tegangan tinggi adalah PEF (Pulsed Electric Field) di mana untuk membunuh mikroba, metode ini menggunakan medan listrik yang dibentuk pulsa. Parameter yang dipelajari pengaruhnya untuk membunuh mikroba dalam pekerjaan ini adalah tegangan, lama perlakuan, dan polaritas. Dengan menggunakan modul Arduino, lama perlakuan dan polaritas tegangan dapat diatur sesuai dengan keinginan. Modul Arduino dihubungkan pada rangkaian bertegangan rendah lalu dengan menggunakan IR LED dan phototransistor dapat mengontrol saklar elektronik H-Bridge yang terisolasi secara galvanik. Tegangan maksimal pada rangkaian H-Bridge adalah 1,3 kV karena kapasitas MOSFET. Maka dari itu dibutuhkan modifikasi pada rangkaian pembangkit tegangan tinggi agar tidak merusak MOSFET. Percobaan dilakukan dengan menghubungkan 8 buah rangkaian tegangan tinggi secara seri 5698,97 Vrms yang memiliki bentuk gelombang bipolar square wave dengan frekuensi 454,5 Hz selama waktu efektif 0,038 detik (durasi perlakuan). Percobaan ini dilakukan pada susu yang baru diperah dari peternakan di daerah Bogor. Susu yang telah mendapat perlakuan dianalisis dengan metode TPC (Total Plate Count). Derajat kematian dari metode TPC menggunakan media PCA (Plate Count Agar untuk mengukur seluruh bakteri) pada pengenceran 10-3 dengan dua sampel (duplo) yang diberi perlakuan PEF dengan dosis sebesar 3,87 x 106 Vrms2detik/cm2 adalah TBUD (Tidak Bisa Untuk Dihitung). Untuk pengenceran 10-4, derajat kematian yang dihasilkan dengan perlakuan yang sama adalah 54,34%. Derajat kematian menggunakan media VRBA (Violet Red Bile Agar untuk mengukur bakteri coliform) pada pengenceran 10-3 pada sampel yang diberi perlakuan PEF dengan dosis 3,87 x 106 Vrms2detik/cm2 adalah 89,81%. Untuk pengenceran 10-4, derajat kematian yang dihasilkan dengan perlakuan yang sama adalah 61,76%. Pada semua percobaan sampel kontrol diambil dari susu dari kemasan yang sama tapi tidak diberi perlakuan. Hasil ini menunjukkan bahwa perlakuan PEF membuat sebagian bakteri akan mati.
