Abstract

Bahan alam merupakan suatu potensi yang banyak dieksplorasi untuk menemukan senyawa yang memiliki khasiat antimikroba, salah satu tanaman yang memiliki aktivitas antibakteri adalah daun papaya jepang (Cnidoscolus aconitifolius (Mill.) I.M. Johnst). Daun pepaya jepang dimaserasi dengan pelarut etanol 96% selama 3x24 jam dan menghasilkan rendemen ekstrak sebanyak 10,81%. Aktivitas antibakteri ditunjukkan pada konsentrasi ekstrak daun pepaya jepang 10000ppm. Pengembangan formulasi sediaan gel dibuat menjadi 4 formula dengan basis yang berbeda yaitu formulasi 1 (HPMC 0,75%); formulasi 2 (HPMC 1%); formulasi 3 (CMC Na 4,5%) dan formulasi 4 (CMC Na 6%). Sediaan gel dilakukan evaluasi yang terdiri dari uji organoleptis, homogenitas, uji pH, daya lekat dan daya sebar. Serta dilakukan uji stabilitas dengan penyimpanan selama 2 minggu 14 hari, yang diamati pada hari ke 7 dan ke 14. Berdasarkan hasil penelitian diperoleh bahwa gelling agent sangat berpengaruh terhadap daya sebar dan daya lekat yang dihasilkan. HPMC dengan konsentrasi yang digunakan sebagai basis menghasilkan daya sebar yang lebih baik dibanding dengan CMC Na karena CMC Na memiliki viskositas yang lebih solid dibanding HPMC. Hasil uji Stabilitas selama 2 minggu penyimpanan menunjukkan bahwa formulasi dengan gelling agent HPMC ialah sediaan yang stabil selama penyimpanan, sedangkan formulasi dengan gelling agent CMC Na menunjukkan ketidakstabilan. / Natural material is a potential that is widely explored to find compounds that have antimicrobial properties, one of the plants that has antibacterial activity is Japanese papaya leaves (Cnidoscolus aconitifolius (Mill.) IMJOHNST) Japanese papaya leaves were macerated with 96% ethanol for 3x24 hours and the totalextract yield is 10.81%. Antibacterial activity was shown at a concentration of 10000 ppm Japanese papaya leaf extract. The development of the gel dosage formulation was made into 4 formulations, namely formulation 1 (HPMC 0.75%); formulation 2 (HPMC 1%); formulation 3 (CMC Na 4.5%) and formulation 4 (CMC Na 6%). The gel preparation was evaluated consisting of organoleptic test, homogeneity, pH test, adhesion, and dispersion power. The stability test was also carried out by storing for 2 weeks which was observed on the 7th and 14th day. Based on the results of the study, it was found that gelling agent had significant effect on spreadability and adhesion of the gel. HPMC in concentration used as the basis showed better spreadability compared with CMC Na because the CMC Na basis had higher viscosity than HPMC. Result from 2 weeks stability test showed that the formulation with HPMC gelling agent showed to be stable during storage, while the formulation with CMC Na gelling agent CMC Na showed to be unstable.
