Abstract

Diabetes melitus merupakan penyakit kelainan metabolik yang ditandai dengan keadaan dimana kadar glukosa darah yang melebihi batas normal. Secara tradisional, daun Pepaya Jepang (Cnidosclus aconitifolius (Mill.) I.M.Johnst.) sering digunakan sebagai pengobatan diabetes, mengobati kanker, penurunan berat badan, mengatasi tekanan darah tinggi dan penyakit ginjal. Penelitian ini bertujuan untuk menguji efektivitas dan menetapkan dosis paling efektif ekstrak etanol 96% daun Pepaya Jepang sebagai antihiperglikemik pada mencit jantan (Mus musculus) yang diinduksi larutan glukosa 5%. Digunakan mencit jantan berjumlah 20 ekor mencit yang dibagi dalam 5 kelompok perlakuan yaitu kelompok kontrol negatif diberi aquadest, kelompok kontrol positif diberi metformin dengan dosis 1,3 mg/20gBBmencit, kelompok perlakuan ekstrak etanol 96% daun Pepaya Jepang dengan dosis 100; 300; 500 mg/20gBBmencit. Tiap kelompok uji dilakukan pengecekkan kadar glukosa darah pada menit 30, 60 dan 90. Hasil rendemen ekstrak yang diperoleh sebesar 10,81%, mengandung senyawa metabolit sekunder diantaranya flavonoid, alkaloid, tanin, saponin, fenol, dan steroid. Ekstrak etanol 96% daun Pepaya Jepang memiliki efektivitas dalam menurunkan kadar glukosa darah pada mencit yaitu pada dosis 100 mg dan dosis 300 mg. / Diabetes mellitus is a metabolic disorder disease defined by blood glucose levels that are higher than normal. Daun Pepaya Jepang (Cnidosclus aconitifolius (Mill.) I.M.Johnst.) have traditionally been used to cure diabetes, cancer, weight reduction, high blood pressure, and kidney illness. The purpose of this study is to establish the most effective dose of 96% ethanol extract of daun Pepaya Jepang as an antihyperglycemic agent in male mice (Mus musculus) caused by a 5 percent glucose solution. Twenty male mice were divided into five treatment groups: the negative control group received aqua dest, the positive control group received metformin at a dose of 1.3 mg/20gBB mice, and the treatment group received a 96 percent ethanol extract of daun Pepaya Jepang at doses of 100, 300, and 500 mg/kg BW mice. Blood glucose levels in each test group were measured at 30, 60, and 90 minutes. The extract yielded 10.81 percent and contained secondary metabolites such as flavonoids, alkaloids, tannins, saponins, phenols, and steroids. At 100 mg and 300 mg doses, a 96 percent ethanol extract of daun Pepaya Jepang was effective in decreasing blood glucose levels in rats
