Abstract

Penyakit kardiovaskular merupakan salah satu penyebab utama kematian pada zaman ini karena adanya gaya hidup yang tidak sehat. Ada tiga kategori umum pengobatan yang biasanya digunakan, yaitu antikoagulan, agen fibrinolitik dan obat antiplatelet. Namun, penggunaan obat-obatan tersebut dapat memberikan efek samping yang berbahaya bagi tubuh seperti pendarahan yang berlebihan. Oleh karena itu, studi tentang penggunaan tanaman herbal, yaitu pare (Momordica charantia) sebagai agen fibrinolitik yang lebih aman perlu dilakukan. Penelitian ini dilakukan mulai dari ekstraksi protease serin dan dilanjutkan dengan uji protease, uji degradasi gumpalan darah, uji hemolitik, dan uji aktivitas fibrinolitik yang dilakukan dengan fibrin plate assay terhadap ekstrak biji, buah, daun dan batang Momordica charantia. Keempat sampel yang digunakan, diketahui mampu mendegradasi gumpalan darah. Selain itu, keempat sampel juga mampu mendegradasi fibrin secara langsung dan dapat diidentifikasikan sebagai γ-hemolisis, karena tidak dapat memecah sel darah merah. Berdasarkan penelitian ini, ekstrak biji pare mempunyai aktivitas fibrinolitik paling tinggi, yang diikuti dengan ekstrak daun pare, ekstrak daging pare dan ekstrak batang pare yang mempunyai aktivitas paling rendah. / Nowadays, cardiovascular disease is one of the main causes of death due to an unhealthy lifestyle. There are three general categories of medications that are commonly used, anticoagulants, fibrinolytic agents and antiplatelet drugs. However, the use of these drugs can provide harmful side effects to the body, such as excessive bleeding. Therefore, studies on the use of herbal plants, like bitter gourd (Momordica charantia) as an alternative for fibrinolytic agents that are safer, need to be carried out. This research was carried out from the extraction of serine proteases and continued with protease assay, blood clot degradation assay, haemolytic tests, and fibrinolytic activity by fibrin plate assay on extracts of seeds, fruits, leaves and stems of Momordica charantia. The four samples used were known to be able to degrade blood clots. In addition, the four samples were also able to degrade fibrin directly and can be identified as γ-hemolysis, because it cannot hydrolyse red blood cells. Based on this study, bitter gourd seed extract had the highest fibrinolytic activity, followed by bitter gourd leaf extract, bitter gourd flesh extract and bitter gourd stem extract which had the lowest activity.
