Abstract

Penggumpalan darah akut, seperti infark miokard dan stroke iskemik, merupakan penyebab utama morbiditas dan mortalitas di seluruh dunia. Namun, beberapa agen antitrombotik yang umum digunakan untuk mengatasinya masih memiliki beberapa kelemahan, sehingga studi ini berfokus terhadap agen alternatif antitrombotik yang lebih aman dan efisien. Pare (Momordica charantia L) yang diketahui memiliki rasa pahit, juga memiliki fungsi dan kegunaan yang erat dikaitkan dengan komponen asam fenolatnya, salah satunya asam galat. Komponen ini merupakan antioksidan dan komponen natural yang memiliki potensi sebagai antitrombotik, serta aman digunakan untuk proses pencegahan gumpalan darah. Oleh karena itu, penelitian ini dilakukan untuk mengevaluasi potensi berbagai sampel pare sebagai agen antitrombotik. Metode untuk penelitian ini adalah uji pembentukan gumpalan darah dan uji aktivitas antitrombotik menggunakan platelet-rich plasma (PRP). Keempat sampel pare melalui kedua uji mampu menghasilkan aktivitas antitrombotik. Hal ini diakibatkan karena asam galat yang terkandung dalam pare mampu menghambat kerja ADP, sehingga proses agregasi trombosit dilemahkan. Dari kedua uji yang dilakukan, aktivitas antritrombotik tertinggi dimiliki oleh biji pare, dengan nilai densitas jumlah sel darah merah yang terlepas adalah 1,136 ± 0,02480 x 108 sel/mL serta persen inhibisi agregasi trombosit sebesar 45,691 ± 0,8342%, diikuti dengan daun, daging buah, dan batang pare. Data kualitatif dan analisis wavelength scan juga menunjukan aktivitas antritrombotik dimiliki oleh sampel biji pare. Oleh karena itu, biji pare memiliki potensi tertinggi sebagai agen antitrombotik./Acute blood clots, such as myocardial infarction and ischemic stroke, are one of the major cause of morbidity and mortality worldwide. However, some commonly used antiplatelet agents still have some weaknesses, so this study focused on finding alternative antiplatelet agents that are safer and more efficient. Bitter gourd (Momordica charantia L.) which is known to have a bitter taste, has functions and uses that are closely related to its phenolic acid components, one of which is gallic acid. This component is an antioxidant and natural component that has potential as an antiplatelet and is safe to use for the process of preventing blood clots. Therefore, this study was conducted to evaluate the potential of various bitter gourd samples as antiplatelet agents. The methods for this study were blood clot formation test and antiplatelet activity test using platelet-rich plasma (PRP). Stem, seed, fruit, and leaves of bitter gourd through both tests were able to produce antiplatelet activity. This is due to the gallic acid contained in bitter gourd that is able to inhibit the function of ADP, so that the platelet aggregation process is attenuated. From the two tests carried out, the highest antithrombotic activity was possessed by bitter gourd seeds, with a density value of red blood cells is 1,136 ± 0,02480 x 108 cells/mL and the percent inhibition of platelet aggregation is 45,691 ± 0,8342% followed by leaves, fruits, and stems of bitter melon. Qualitative data and analysis of wavelength scan also confirmed antitplatelet activity of bitter gourd seeds. Therefore, bitter melon seeds have the highest potential as an antiplatelet agent.
