Abstract

Methylene blue is one of the most used synthetic dye. However, the excessive application of methylene blue are proven to result in undesired effect, such as cyanosis, methaemoglobinaemia, formation of Heinz bodies, and skin discoloration. This research aimed to evaluate the ability of Pelita Harapan University bacteria collections on decolorizing methylene blue on rice bran medium. Pseudomonas aeruginosa (KMB 2) exhibited the ability to thrive on rice bran supplemented with methylene blue. It showed considerable decolorization of methylene blue (21.31 %) when incubated on room temperature (25 ºC) with pH 6.69. Semi purification were applied on crude enzyme by acetone precipitation. Three bands were produced on SDS-PAGE, predicted as lignolytic enzyme (56.18 kDa), FMN-dependent NADHazoreductase (17.65 kDa), and ring-cleaving dioxygenase (11.20 kDa). Along the decolorization process of methylene blue, secondary metabolites produced were analyzed and reflected the formation of leucomethylene blue and decolorization of thionolin./ Methylene blue merupakan salah satu pewarna sintetis yang banyak dimanfaatkan. Pengaplikasian dari pewarna ini secara berlebihan dapat menyebabkan dampak berbahaya, seperti sianosis, methaemoglobulinemia, pembentukkan badan Heinz, dan diskolorisasi kulit. Penelitian ini dilakukan dengan tujuan untuk mengevaluasi kemampuan bakteri koleksi UPH yang mampu mendekolorisasi methylene blue dalam medium bekatul. Pseudomonas aeruginosa (KMB 2) merupakan isolat bakteri yang mampu tumbuh dan melakukan dekolorisasi pada medium bekatul yang disuplementasi dengan methylene blue. Bakteri ini menunjukkan persentase dekolorisasi yang relatif tinggi (21,31 %) ketika diinkubasi pada suhu ruang (25 oC) pada pH 6,69. Pada pengujian enzim, semipurifikasi enzim dibuat dengan mempresipitasi crude enzyme dengan aseton. Hasil presipitasi enzim menunjukkan terbentuknya tiga pita protein pada SDS-PAGE yang diprediksi yang diprediksi merupakan enzim lignolitik (56,18 kDa), FMN-dependent NADHazoreductase (17,65 kDa), serta ring-cleaving dioxygenase (11,20 kDa). Metabolit sekunder yang dihasilkan selama proses dekolorisasi methylene blue menunjukkan terbentuknya leucomethylene blue dan terjadinya dekolorisasi thionolin.
