Abstract

Industri tekstil merupakan salah satu industri yang memiliki peranan penting dalam pertumbuhan ekonomi negara Indonesia, Akan tetapi terdapat juga beberapa dampak negatif yang dihasilkan oleh industri tekstil, salah satunya adalah pencemaran limbah pewarna tekstil ke lingkungan. Metode biologis merupakan metode yang dapat diaplikasikan untuk mengatasi permasalahan tersebut dan memiliki berbagai keuntungan jika dibandingkan dengan pendekatan kimia dan fisika. Terdapat beberapa mikroorganisme (bakteri, fungi, khamir, dan alga) yang memiliki kemampuan dekolorisasi. Isolasi dilakukan dengan metode spreading pada media YPD agar yang telah ditambahkan dengan berbagai pewarna. Isolat khamir memiliki kemampuan dekolorisasi yang baik pada pewarna malachite green dengan nilai rata-rata dekolorisasi 80,4 ± 5,4% selama 3 hari inkubasi pada konsentrasi pewarna 50 ppm. Uji biodegradasi (enzim ekstraseluler) dilakukan dengan menginkubasi CFS hasil dekolorisasi ke dalam pewarna malachite green dengan konsentrasi 6 ppm. Hasil menunjukkan bahwa enzim ekstraseluler tidak berperan dalam proses dekolorisasi oleh isolat khamir. Uji kemampuan bioakumulasi dan bioabsorpsi dilakukan dengan menginkubasi isolat khamir pada media YPD dengan malachite green dengan konsentrasi 5 ppm dan diamati selama 2, 4, 6, dan 12 jam. Hasil menunjukkan bahwa bioabsorpsi dan/atau bioakumulasi berperan pada tahapan awal dekolorisasi yang kemudian diikuti oleh biodegradasi intraseluler. Uji MIC dilakukan pada konsentrasi 10, 20, 50, 100, 200, 500, dan 1000 ppm. Isolat khamir memiliki aktivitas dekolorisasi pada konsentrasi pewarna 10 – 500 ppm dan tidak memiliki aktivitas pada konsentrasi malachite green 1000 ppm. Identifikasi dilakukan dengan pendekatan morfologi dan juga molekuler. Melalui pendekatan morfologi (morfologi koloni serta bentuk sel), diperkirakan isolat merupakan khamir dari genus saccharomyces. Identifikasi molekuler dilakukan dengan menganalisis homologi dan filogenetik. Hasil menunjukkan bahwa isolat khamir asal ragi wine merupakan S. cerevisiae. / The textile industry is one of the important industries that contribute to Indonesia’s economic growth. However, there are some negative effects that textile industry produced. One of which is the pollution by textile dye waste to the environment. Biological approach can be applied to overcome the problems and offer various of advantages compared to chemical dan physical approach. Microorganism (bacteria, fungi, yeast, and algae) has known for their decolorizing activity. The isolation of yeast from wine yeast was carried out by spreading method on YPG agar that have been added with dyes. Wine yeast isolate has good decolorization activity on malachite green with an average decolorization value of 80.4 ± 5.4% after 3 days of incubation at a dye concentration of 50 ppm. Extracellular Biodegradation test done by incubating CFS from the previous decolorization process into malachite green at a concentration of 6 ppm. Result shows that extracellular enzyme does not play a role in the decolorization process. Bioaccumulation and bioabsorption test done by incubating wine yeast isolate on YPD broth that have been added with malachite green with a dye concentration of 6 ppm and observed for 2, 4, 6, and 12 hours. Result shows that bioabsorption and/or bioaccumulation take a part in the early stage of decolorization process followed by intracellular biodegradation. MIC test done at dye concentration of 10, 20, 50, 100, 200, 500, and 1000 ppm. Yeast isolate has decolorizing activity on dye concentration 10 – 500 ppm and does not decolorize at dye concentration 1000 ppm. Yeast identification done by morphology and molecular approach. Morphology identification shown that yeast isolate estimated to be Saccharomyces. Molecular identification done by analyzing the homology and phylogenetic of the yeast. Result show that wine yeast isolate is S. cerevisiae.
