Abstract

Prebiotik adalah senyawa oligosakarida yang termasuk dalam bahan pangan yang tidak dapat dicerna oleh enzim tubuh manusia. Prebiotik secara selektif menstimulasi pertumbuhan dan aktivitas probiotik, terutama pada genus bakteri probiotik seperti Lactobacillus sp dan Bifidobacterium sp. Bifidobacterium sp banyak ditemukan pada feses bayi yang mengonsumsi ASI. Penelitian ini bertujuan untuk menguji pertumbuhan dan kemampuan isolat BR5 dan BR6 dalam memfermentasi berbagai substrat prebiotik dengan menggunakan uji aktivitas fermentasi, uji enumerasi, dan perbedaan morfologi sel. Metode penelitian terdiri dari uji aktivitas fermentasi pada isolat BR5 dan BR6 sebagai uji kualitatif yang ditumbuhkan pada medium TPY dan MRS dengan penambahan masing-masing 2% FOS, GOS, inulin, IMO, XOS, dan glukosa sebagai kontrol positif, serta medium tanpa gula sebagai kontrol negatif dengan menggunakan indikator fermentasi bromocresol purple dan diinkubasi selama 24 jam pada suhu 37°C secara anaerobik obligat; uji enumerasi pada isolat BR5 dan BR6 sebagai uji kuantitatif dilakukan seperti pada uji kualitatif tetapi diinkubasi selama 48 jam; pengamatan morfologi sel dilakukan dengan gram staining yang ditumbuhkan pada medium TPY dan MRS dengan penambahan berbagai prebiotik. Hasil penelitian menunjukkan, pada uji kualitatif isolat BR5 hanya dapat memfermentasi FOS, dan isolat BR6 hanya pada medium TPY dapat memfermentasi semua prebiotik kecuali XOS. Uji enumerasi isolat BR5 pada kedua medium yang digunakan menunjang hasil uji kualitatif sedangkan pada isolat BR6 tidak sesuai dengan hasil uji kualitatif. Pengamatan morfologi sel tidak menunjukkan adanya perbedaan morfologi sel dengan menggunakan substrat prebiotik. / Prebiotics are oligosaccharide compounds that are included in food ingredients that cannot be digested by the enzymes of the human body. Prebiotics selectively stimulate the growth and activity of probiotics, especially in probiotic bacterial genera such as Lactobacillus sp and Bifidobacterium sp. Bifidobacterium sp is found in the feces of infants who consume breast milk. This study aimed to test the growth and ability of BR5 and BR6 isolates in fermenting various prebiotic substrates using fermentation activity tests, enumeration tests, and differences in cell morphology. The research method consisted of a fermentation activity test on isolates BR5 and BR6 as a qualitative test grown on TPY and MRS medium with the addition of 2% FOS, GOS, inulin, IMO, XOS, and glucose as a positive control, and a medium without sugar as a negative control using bromocresol purple fermentation indicator and incubated for 24 hours at 37°C obligate anaerobically; enumeration test on isolates BR5 and BR6 as a quantitative test was carried out as in the qualitative test but incubated for 48 hours; cell morphology observations were carried out by gram staining grown on TPY and MRS medium with the addition of various prebiotics. The results showed that in the qualitative test, BR5 isolates could only ferment FOS, and BR6 isolates only on TPY medium could ferment all prebiotics except XOS. The enumeration test for BR5 isolates in the two mediums used supported the qualitative test results, while the BR6 isolates did not match the qualitative test results. Cell morphology observations did not show any differences in cell morphology using prebiotic substrates.
