Abstract

Radikal bebas merupakan molekul yang sangat reaktif karena mengandung satu atau lebih dari satu elektron yang tidak saling berpasangan. Radikal bebas yang dihasilkan secara terus menerus akan menyebabkan terjadinya kerusakan fungsi sel-sel tubuh yang menjadi pemicu berbagai macam penyakit. Antioksidan dapat menghambat terjadinya pembentukan radikal bebas didalam tubuh dengan menjadi pendonor elektron untuk radikal bebas. Salah satu tumbuhan yang diduga berpotensi sebagai antioksidan yaitu tumbuhan tungurrut (C. tungurrut). Penelitian ini bertujuan untuk mengetahui kandungan senyawa metabolit sekunder, mengetahui kadar enolik dan flavonoid total, dan aktivitas antioksidan ekstrak etanol 70% daun tungurrut (C. tungurrut) dengan metode DPPH yang dinyatakan dalam parameter IC50 (Inhibition concentration 50%). Serbuk simplisia daun tungurrut di ekstrak menggunakan pelarut 70% dengan metode maserasi dan dilakukan skrining fitokimia, dilanjutkan dengan uji aktivitas antioksidan menggunakan metode DPPH, serta penetapan kadar fenolik menggunakan asam galat sebagai larutan strandar dengan metode folin ciocalteu dan flavonoid total dengan metode kolorimetri dengan AlCl3 sebagai pereaksi dan kuersetin sebagai larutan standar. Dari proses ekstraksi diperoleh rendamen sebesar 9,4107% b/b. Hasil dari skrining fitokimia ekstrak mengandung senyawa alkaloid, flavonoid, saponin, tanin, fenol dan steroid. Hasil uji aktivitas antioksidan secara kuantitatif menunjukkan bahwa ektrak etanol 70% daun tungurrut (C. tungurrut) memiliki nilai IC50 sebesar 36,86 ppm. Kadar fenolik total yang diperoleh yaitu sebesar 1109,3497±235,4152 mg GAE/g ekstrak. Kadar flavonoid total yang diperoleh yaitu sebesar yaitu sebesar 22,4413 ± 3,6796 mg QE/g ekstrak. Berdasarkan hasil yang diperoleh menggunakan parameter IC50 ekstrak etanol 70% daun tungurrut (C. tungurrut) memiliki aktivitas antioksidan dengan kategori sangat kuat./Free radicals are highly reactive molecules because they contain one or more unpaired electrons. Free radicals that are produced continuously will cause damage to the function of body cells that trigger various diseases. Antioxidants can inhibit the formation of free radicals in the body by becoming electron donors for free radicals. One of the plants that are thought to have potential as antioxidants is the tungurrut plant (C. tungurrut). This study aims to determine the content of secondary metabolites, to determine the total enolic and flavonoid levels, and the antioxidant activity of 70% ethanol extract of tungurrut (C. tungurrut) leaves using the DPPH method which is stated in the IC50 parameter (Inhibition concentration 50%). Simplicia powder of tungurrut leaf was extracted using 70% solvent by maceration method and phytochemical screening was carried out, followed by antioxidant activity test using the DPPH method, and determination of phenolic levels using gallic acid as standard solution using the Folin ciocalteu method and total flavonoids by colorimetric method with AlCl3 as reagent and quercetin as standard solution. From the extraction process, the yield was 9.4107% w/w. The results of phytochemical screening of extracts contained alkaloids, flavonoids, saponins, tannins, phenols and steroids. The results of the antioxidant activity test quantitatively showed that the 70% ethanol extract of tungurrut (C. tungurrut) leaves had an IC50 value of 36.86 ppm. The total phenol content obtained was 1109.3497±235.4152 mg GAE/g extract. The total flavonoid content obtained was 22.4413 ± 3.6796 mg QE/g extract. Based on the results obtained using the IC50 parameter, 70% ethanol extract of tungurrut (C. tungurrut) leaves has antioxidant activity with a very strong category.
