Abstract

Karsinoma payudara merupakan jenis karsinoma terbanyak di Indonesia. Amplifikasi Human Epidermal Growth Factor Receptor 2 (HER2) digunakan untuk menentukan terapi yang akan diterima. Data mengenai hubungan aspek klinikopatologi dan amplifiksi HER2 menggunakan Dual-In Situ Hybridization (DISH) pada karsinoma payudara masih sedikit, dan hasil yang diperoleh tidak konsisten. Penelitian ini bertujuan untuk mengetahui hubungan antara aspek klinikopatologi dengan amplifikasi HER2 menggunakan DISH pada karsinoma payudara. Penelitian ini menggunakan metode studi potong lintang dengan jenis penelitian analitik komparatif kategorik tidak berpasangan. Populasi sampel pada penelitian ini adalah pasien karsinoma payudara di Rumah sakit MRCCC Siloam pada tahun 2019-2022. Pengambilan data dilakukan dengan metode sampel purposif, dan secara retrospektif menggunakan data sekunder, yaitu rekam medis. Data yang diperoleh akan diolah menggunakan program SPSS 25.0 dan analisa data menggunakan metode Chi Square atau Uji Fisher dan uji T-test atau Mann Whitney-U. Analisis bermakna jika p value < 0.05. Hasil penelitian yang didapatkan adalah tidak terdapat hubungan antara aspek klinikopatologi dengan amplifikasi HER2 berdasarkan DISH dengan p-value >0,05. Amplifikasi HER2 yang positif lebih banyak didapatkan pada ukuran tumor besar dan grading tumor tinggi. Tidak terdapat hubungan signifikan antara usia, lokasi tumor, ukuran tumor, grading tumor, invasi limfovaskular, metastasis KGB, ekspresi ER, ekspresi PR dengan amplifikasi HER2 berdasarkan DISH pada pasien karsinoma payudara dengan ekspresi HER2 +2 di Rumah Sakit MRCCC Siloam Semanggi. / Breast carcinoma is the most common type of carcinoma in Indonesia. Human Epidermal Growth Factor Receptor 2 (HER2) amplification is used to determine which therapy will be done. There are only a few data regarding the relationship between clinicopathological aspects and HER2 amplification using Dual-In Situ Hybridization (DISH) in breast carcinoma with inconsistent result. This study aims to determine the relationship between clinicopathological aspects and the amplification of HER2 using DISH in breast carcinoma. This study uses a cross-sectional method which is categorized as an unpaired categorical comparative analytic study. The sample population for this study are breast carcinoma patients at MRCCC Siloam Hospital in 2019-2022. Data are collected retrospectively using purposive sampling method, namely medical records. The data obtained will be processed using the SPSS 25.0 program and data analysis will be done using the Chi Square method or Fisher’s exact test and T-test or Mann Whitney-U. The analysis is considered meaningful if the p value is <0.05. The results of the study showed that there was no relationship between clinicopathological aspects and HER2 amplification based on DISH with p-value > 0.05. Positive HER2 amplification was found more in large tumor size and high tumor grade. There is no significant relationship between age, tumor location, tumor size, tumor grading, lymphovascular invasion, lymph node metastasis, ER expression, PR expression with HER2 amplification based on DISH in breast carcinoma patients with HER2 +2 expression at MRCCC Siloam Semanggi Hospital.
