Abstract

Keberadaan radikal bebas sebagai hasil metabolisme sel normal pada tubuh manusia berperan sebagai pertahanan tubuh. Dalam keadaan jumlah radikal bebas yang berlebih berakibat pada kerusakan sel dan hal ini disebut stres oksidatif. Keadaan stress oksidatif mampu memicu timbulnya penyakit bersifat kronik dan akumulatif. Antioksidan adalah senyawa yang mampu meredam proses oksidasi akibat radikal bebas, dengan mekanisme menghambat proses oksidasi. Salah satu sumber antioksidan yaitu hasil metabolit sekunder tumbuhan. Tumbuhan C. argentea merupakan tumbuhan yang diduga terdapat aktivitas antioksidan dengan kandungan senyawa fenol dan flavonoid. Penelitian ini ditujukan untuk mengetahui kandungan senyawa metabolit sekunder, total fenol dan flavonoid, dan aktivitas antioksidan ekstrak batang etil asetat dan etanol 70% C. argentea menggunakan metode DPPH dan FRAP. Total fenolik ekstrak batang C. argentea etanol diperoleh 471,8036 ± 27.0800 mg GAE/g ekstrak dan ekstrak batang C. argentea etil asetat diperoleh 165,7678 ± 6,2483 mg GAE/g. Total flavonoid ekstrak batang C. argentea etanol diperoleh 22,6989 ± 0,3373 mg QE/g dan ekstrak etil asetat 27,5076 ± 0,7506 mg QE/g. Senyawa fenolik dalam pengujian ini berkontribusi besar sebagai agen antioksidan pada ekstrak batang C. argentea etanol. Berdasarkan hasil pengujian baik pada ekstrak batang C. argentea etanol maupun etil asetat memiliki faktor dominan yang berbeda sebagai agen antioksidannya./The existence of free radicals as a result of metabolism of normal cells in the human body has a role as body defense. In a state of excess free radicals result in cell damage and this is referred to as oxidative stress. The state of oxidative stress can trigger chronic and accumulative diseases. Antioxidants are compounds that are able to reduce the oxidation process due to free radicals, with the mechanism of inhibiting the oxidation process. One source of antioxidants is the result of plant secondary metabolites.plant C. argentea that is thought to have antioxidant activity with phenol and flavonoid compounds. This study was aimed to determine the content of secondary metabolites, total phenols and flavonoids, and antioxidant activity of ethyl acetate and 70% ethanol stem extracts of C. argentea using DPPH and FRAP methods. Total phenolic stem extract of C. argentea ethanol obtained 471.8036 ± 27.0800 mg GAE/g extract and stem extract C. argentea ethyl acetate obtained 165.7678 ± 6.2483 mg GAE/g. The total flavonoids of C. argentea stem extract ethanol obtained 22.6989 ± 0.3373 mg QE/g and ethyl acetate extract 27.5076 ± 0.7506 mg QE/g. The phenolic compounds in this test contributed greatly as antioxidant agents to the ethanol stem extract of C. argentea but did not contribute greatly to the ethyl acetate extract. Based on the test results, both ethanol and ethyl acetate stem extracts of C. argentea have different dominant factors as antioxidant agents.
