Abstract

Limbah pewarna tekstil telah menjadi salah satu masalah dalam ekosistem perairan. Hal tersebut dikarenakan sebagian besar limbah tersebut sering dibuang ke perairan sehingga dapat mengakibatkan penghambatan pertumbuhan tumbuhan, mutagenitas, menganggu proses sintesis tumbuhan, dan lain sebagainya. Oleh karena itu, diperlukan solusi untuk menangani permasalahan ini yaitu dengan menggunakan mikroba untuk mendekolorisasikan limbah pewarna tekstil tersebut menjadi molekul yang lebih aman untuk lingkungan. Pada penelitian ini, dilakukan karakterisasi dan identifikasi mikroba yang diisolasi dari limbah pabrik batik. Tahapan penelitian ini meliputi: (1) Isolasi sampel dari limbah pabrik batik, (2) Pemurnian sampel yang telah diisolasi dari limbah pabrik batik, (3) Identifikasi sampel secara molekuler, (4) Menguji kemampuan dekolorisasi sampel diberbagai warna, (5) Menguji kemampuan dekolorisasi sampel diberbagai konsentrasi, (6) Menguji kemampuan dekolorisasi sampel secara enzimatik, (7) Mengidentifikasi keberadaan enzim pendekolorisasi sampel. Hasil yang diperoleh menunjukan bahwa mikroba yang diisolasi dari limbah pabrik batik adalah bakteri Alcaligenes sp. yang memiliki kemampuan dekolorisasi terbaik dipewarna remazol red RB dan dapat mendekolorisasikan konsentrasi 100, 200, 300, 400, 500 ppm sebesar 70% dalam waktu 4 hari. Hasil yang diperoleh mengindikasikan bahwa Alcaligenes sp. dapat mendekolorisasikan pewarna secara enzimatik atau ekstraseluler dan enzim yang berperan dalam dekolorisasi kemungkinan besar adalah azoreductase. / Textile dye waste has become a problem in aquatic ecosystems. This is because most of the waste is often disposed of into the waters so that it can inhibit plant growth, mutagenicity, disrupt plant synthesis processes, and so on. Therefore, a solution is needed to deal with this problem, namely by using microbes to decolorize the textile dye waste into molecules that are safer for the environment. In this study, the characterization and assistance of microbes isolated from batik factory waste were carried out. The stages of this research included: (1) Isolation of samples from batik factory waste, (2) Purification samples that had been isolated from batik factory waste, (3) Molecular identification of the sample, (4) Test of the ability to decolorize the sample in various colors, (5) Test the ability of the sample to decolorize in various concentrations, (6) Test the ability of the sample to decolorize enzymatically, (7) Identify the presence of enzymes to decolorize the sample. The results showed that the microbes isolated from batik factory waste were Alcaligenes sp. bacteria which had the best decolorization ability in remazol red RB and could decolorize concentrations of 100, 200, 300, 400, 500 ppm by 70% within 4 days. The results obtained indicate that Alcaligenes sp. can decolorize dyes enzymatically or extracellularly and the enzyme that plays a role in decolorization is most likely azoreductase.
