Abstract

Bifidobacterium breve sering digunakan sebagai bakteri probiotik dan ditemukan di dalam saluran pencernaan manusia serta air susu ibu. Sebagai probiotik, B. breve memiliki kemampuan adhesi pada mukus dan sel epitel usus, sehingga berperan penting dalam kolonisasi di saluran pencernaan. Pada penelitian ini, dilakukan karakterisasi sifat probiotik B. breve BS2-PB5 yaitu melalui analisis kemampuan adhesi pada model mukus babi dan analisis respon terhadap stres oksidatif (aerobik dan mikroaerofilik) terhadap viabilitas sel. Percobaan uji respon terhadap stres oksidatif BS2-PB5 pada kondisi aerobik dan mikroaerofilik dilakukan melalui metode colony-forming unit. Percobaan uji kemampuan adhesi BS2-PB5 dilakukan melalui metode adherence assay mengggunakan model mukus babi tipe II dan BSA sebagai kontrol yang diimobilisasi pada microplates untuk selanjutnya dihitung persentase adhesinya. Hasil analisis respon terhadap stres oksidatif pada BS2-PB5 di tiga kondisi berbeda, aerobik, mikroaerofilik, dan anaerobik masing-masing memiliki rata-rata viabilitas sel, yaitu 2,63 ± 0,67 CFU/mL; 2,70 ± 0,56 CFU/mL; dan 3,73 ± 0,73 CFU/mL, menunjukkan viabilitas sel pada BS2-PB5 paling tinggi hingga paling rendah adalah pada kondisi anaerobik, mikroaerofilik, dan aerobik. Hasil analisis kemampuan adhesi BS2-PB5 pada model mukus babi dan BSA menunjukkan setelah diinkubasi selama 1 jam, 2 jam, dan 3 jam pada suhu 37°C secara anaerobik masing-masing untuk mukus babi adalah 0,41 ± 0,22 CFU/mL; 0,63 ± 0,10 CFU/mL; dan 1,01 ± 0,13 CFU/mL; untuk BSA adalah 0,26 ± 0,09 CFU/mL; 0,47 ± 0,31 CFU/mL; dan 1,05 ± 0,55 CFU/mL. Persentase adhesi pada mukus babi dan BSA yang diinkubasi selama 1 jam, 2 jam, dan 3 jam memiliki persentase adhesi, untuk mukus babi masing-masing adalah 83,59%; 86,4%; dan 89,01%; untuk BSA adalah 81,34%; 83,99%; dan 88,69%. Persentase adhesi dan jumlah BS2-PB5 yang menempel pada mukus babi dan BSA memiliki nilai rata-rata paling tinggi hingga paling rendah adalah masa inkubasi selama 3 jam, 2 jam, dan 1 jam. / Bifidobacterium breve is usually used as a probiotic bacteria that is found in the human digestive tract and breast milk. As a probiotic, B. breve has the ability to adhere to mucus and intestinal epithelial cells, thereby playing an important role in colonization in the digestive tract. In this study, the probiotic properties of B. breve BS2-PB5 were determined by analyzing the adhesion ability of the porcine mucus model and the response to oxidative stress (aerobic and microaerophilic) on cell viability. The response test to BS2-PB5 oxidative stress under aerobic and microaerophilic conditions was carried out using the colony-forming unit method. The BS2-PB5 adhesion test was carried out using the adherence assay method using a type II porcine mucus model and BSA as a control to immobilize on microplates to further determine the percentage of adhesion. The results of the analysis of the response to oxidative stress on BS2-PB5 had an average cell viability of 3.73 ± 0.73 CFU/mL; 2.70 ± 0.56 CFU/mL; and 2.63 ± 0.67 CFU/mL, indicating the highest to lowest cell viability under anaerobic, microaerophilic, and aerobic conditions. The analysis of the adhesion capacity of BS2-PB5 in porcine mucus and BSA revealed that, after incubation for 1 hour, 2 hours, and 3 hours at 37°C, anaerobically, the corresponding values for porcine mucus were 0.41 ± 0.22 CFU/mL; 0.63 ± 0.10 CFU/mL; and 1.01 ± 0.13 CFU/mL, whereas, for BSA were 0.26 ± 0.09 CFU/mL; 0.47 ± 0.31 CFU/mL; and 1.05 ± 0.55 CFU/mL. The percentage of adhesion to porcine mucus and BSA after incubation for 1 hour, 2 hours, and 3 hours was 83.59%; 86.4%; and 89.01% for porcine mucus, and 81.34%; 83.99%; and 88.69% for BSA. The incubation times of 3 hours, 2 hours, and 1 hour had the greatest to lowest average values for the percentage of adhesion and the amount of BS2-PB3 attached to the porcine mucus and BSA.
