Abstract

Bifidobacterium animalis subsp. lactis strain BR2-5 yang telah berhasil diisolasi dari feses bayi memiliki potensi untuk digunakan sebagai probiotik. Dalam penelitian ini, dilakukan pengujian untuk mengevaluasi pengaruh dari mikronekapsulasi terhadap viabilitas BR2-5 setelah proses spray drying; selama masa penyimpanan (shelf life); serta setelah melalui uji Simulated Gastric Juice (SGJ) dan garam empedu. Selain itu, dilakukan pula uji in vivo untuk menganalisis pengaruh pemberian BR2-5 hasil mikroenkapsulasi terhadap mencit BALB/c. Metode penelitian yang dilakukan meliputi mikroenkapsulasi BR2-5 dengan gabungan dua matriks berupa 5% skim milk dan 5% maltodekstrin, pengeringan mikrokapsil dengan spray drying pada suhu 130°C; uji viabilitas dan penentuan survival rate BR2-PB5 sebelum dan setelah spray drying; selama 8 minggu masa penyimpan (shelf life) di suhu 4°C dan 25°C; setelah terpapar SGJ dan garam empedu 0,3% selama 3 jam. Pengaruh pemberian pelet hasil suplementasi BR2-5 diamati dari berat feses, jumlah dropping, dan pH dari feses mencit BALB/c. Berdasarkan hasil yang diperoleh, BR2-5 dengan dan tanpa mikroenkapsulasi masing-masing memiliki survival rate sebesar 77,35% dan 52,84%. Shelf life dari mikrokapsul BR2-5 yang disimpan selama 8 minggu pada suhu 4°C dan 25°C tidak mengalami penurunan viabilitas yang signifikan dengan survival rate masing- masing sebesar 88,78% dan 78,80%. Mikroenkapsulasi dapat melindungi BR2-5 dari paparan SGJ dan garam empedu 0,3% selama 3 jam dengan survival rate masing-masing sebesar 42,76% dan 63,42%. Hasil pengamatan in vivo terhadap feses mencit BALB/c selama 3 minggu tidak menunjukkan adanya perubahan signifikan dari berat feses, namun terjadi penambahan jumlah droppingyang signifikan, yaitu sebanyak 14,34%. pH feses mengalami penurunan signifikan dari pH 6 ke pH 5 dimulai dari minggu ke-2. Kesimpulan penelitian: mikroenkapsulasi dapat menjaga viabilitas BR2-5 setelah spray drying, uji shelf life selama 8 minggu di suhu 4°C dan 25°C, uji in vitro dengan SGJ dan garam empedu 0,3%. Pemberian BR2-5 hasil mikroenkapsulasi selama 21 hari dapat memperbanyak jumlah dropping dan menurunkan pH pada feses mencit BALB/c. / Bifidobacterium animalis subsp. lactis strain BR2-5 has been successfully isolated from infant faeces and has the potential to be used as a probiotic. In this study, a test was conducted to evaluate the effect of microencapsulation on the viability of BR2-5 after the spray drying process; during the storage period (shelf life); and after exposure to Simulated Gastric Juice (SGJ) and bile salt. In addition, in vivo tests were also carried out to analyze the effect of microencapsulated BR2-5 on BALB/c mice. The research method used included BR2-5 microencapsulation with a combination of 5% skim milk and 5% maltodextrin; drying the microcapsule by spray drying at 130°C; viability test and determination of BR2-5 survival rate before and after spray drying, after 8 weeks of shelf life at 4°C and 25°C, after exposure to SGJ and 0.3% bile salts for 3 hours. The effect of pellets administration with BR2-5 supplementation was observed through changes in faecal weight, the amount of faeces dropping, and pH of BALB/c mice faeces. Based on the results obtained, BR2-5 with and without microencapsulation had survival rates of 77.35% and 52.84%, respectively. The shelf life of BR2-5 microcapsules stored for 8 weeks at 4°C and 25°C did not experience a significant decrease in viability with survival rates of 88.78% and 78.80%, respectively. Microencapsulation could protect BR25 from exposure to SGJ and 0.3% bile salts for 3 hours with a survival rate of 42.76% and 63.42% respectively. The results of in vivo observations of the faeces of BALB/c mice for 3 weeks did not show any significant changes in the weight of the faeces but there was a significant addition of the amount of dropping, which was 14.34%. The pH of the faeces decreased significantly from pH 6 to pH 5 starting from the second week. The conclusion of the study: microencapsulation can maintain BR2-5 viability after spray drying, shelf life for 8 weeks at 4°C and 25°C, in vitro test with SGJ and 0.3% bile salt. Administration of BR2-5 as a result of microencapsulation for 3 weeks increased the amount of dropping and lowered the pH in the faeces of BALB/c mice.
