Abstract

Tanaman Brotowali (Tinospora crispa L.) memiliki aktivitas sebagai inhibitor α-amilase sehingga glukosa dalam darah akan mengalami penurunan. Enzim α-amilase memiliki peran dalam pembentukkan gula, yaitu mengubah amilosa menjadi monosakarida. Tanaman ini juga memiliki aktivitas sebagai penghambat enzim α-glukosidase, yang mana enzim ini berperan dalam menghidrolisis karbohidrat menjadi gula sederhana (glukosa) pada usus. Oleh karena itu, penelitian ini dilakukan untuk mengetahui dan menganalisis aktivitas ekstrak etil asetat Batang Brotowali (Tinospora crispa (L.) Hook.f. & Thomson) dalam menurunkan kadar glukosa darah pada mencit jantan (Mus musculus) galur Balb/C. Ekstrak Batang Brotowali (Tinospora crispa (L.) Hook.f. & Thomson) diperoleh dengan metode maserasi dengan pelarut etil asetat. Skrining fitokimia dilakukan untuk mengetahui kandungan senyawa, selanjutnya dilakukan uji antihiperglikemik yang diinduksi oleh aloksan terhadap 25 ekor mencit yang terbagi dalam 5 kelompok perlakuan, yaitu kontrol negatif (CMC Na) 1%, kontrol positif (Metformin) 1,3 mg/20 gBB mencit, ekstrak etil asetat Batang Brotowali (Tinospora crispa (L.) Hook.f. & Thomson) dosis 100 mg/KgBB mencit, 300 mg/KgBB mencit, dan 500 mg/KgBB mencit. Hasil yang diperoleh, yaitu % rendemen ekstrak etil asetat batang brotowali sebanyak 6,89% yang mengandung senyawa metabolit sekunder alkaloid,fenol, flavonoid, saponin, tanin, steroid dan quinon. Analisis data dilakukan menggunakan software basis SPSS 26 dengan uji One-Way ANOVA diperoleh hasil terdapat perbedaan signifikan (p < 0,05) yang menyatakan bahwa ekstrak etil asetat batang brotowali memiliki perbedaan yang bermakna dengan kontrol negatif dan memiliki penurunan kadar glukosa darah efektif karena nilai signifikan (p > 0,05) yang tidak berbeda bermakna dengan Metformin. Dosis efektif yaitu 100 mg/KgBB, karena dengan dosis rendah sudah dapat menurunkan kadar glukosa darah yang tidak berbeda bermakna dengan kontrol positif./Brotowali plant (Tinospora crispa L.) has activity as an α-amylase inhibitor so that glucose in the blood will decrease. The α-amylase enzyme has a role in the formation of sugar, namely converting amylose into monosaccharides. This plant also has activity as an inhibitor of the α-glucosidase enzyme, in which this enzyme plays a role in hydrolyzing carbohydrates into simple sugars (glucose) in the intestine. Therefore, this study was conducted to determine and analyze the activity of ethyl acetate extract of Brotowali stem (Tinospora crispa (L.) Hook.f. & Thomson) in reducing blood glucose levels in male mice (Mus musculus). Brotowali Stem Extract (Tinospora crispa (L.) Hook.f. & Thomson) was obtained by maceration method with ethyl acetate solvent. Phytochemical screening was carried out to determine the content of the compounds, then an alloxan-induced antihyperglycemic test was carried out on 25 mice divided into 5 treatment groups, namely negative control (CMC Na) 1%, positive control (Metformin) 1.3 mg/20 g BW mice , Brotowali stem ethyl acetate extract (Tinospora crispa (L.) Hook.f. & Thomson) dose of 100 mg/KgBW mice, 300 mg/KgBW mice, and 500 mg/KgBW mice. The results obtained were 6.89% yield of Ethyl acetate extract of brotowali stems which contained secondary metabolites of alkaloids, phenols, flavonoids, saponins, tannins, steroids and quinones. Data analysis was carried out using the SPSS 26 based software with the One-Way ANOVA test. The results showed that there was a significant difference (p <0.05) which stated that the Ethyl acetate extract of brotowali stems had a significant difference with the negative control and had a decrease in effective blood glucose levels due to the significant (p > 0.05) which was not significantly different from Metformin. The effective dose is 100 mg/KgBW, because low doses can lower blood glucose levels which are not significantly different from positive controls.
