Abstract

Limbah dari logam berat telah menjadi salah satu pencemaran yang harus diperhatikan karena bersifat sangat berbahaya. Hal tersebut dikarenakan logam berat seperti tembaga dapat mempengaruhi lingkungan dan mahluk hidup dengan mudah. Tembaga sendiri dapat digunakan mahluk hidup sebagai mikro-nutrisi, salah satunya menjadi kofaktor, tetapi dalam jumlah besar akan menjadi toksik yang dapat menyebabkan mutasi. Solusi untuk remediasi tembaga di lingkungan sudah dilakukan dengan cara kimia maupun fisika, tetapi cara itu sendiri kurang efektif dan memiliki biaya operasi yang relatif mahal. Oleh sebab itu dibutuhkan cara yang lebih ekonomis serta efektif dengan menggunakan bakteri indigen. Bakteri indigen merupakan bakteri yang diisolasi dari tempat yang tercemar limbah. Pada penelitian ini, dilakukan analisa kemampuan akumulasi dan biosorpsi Pseudomonas aeruginosa D2 diisolasi dari sungai sukolilo terhadap tembaga. Tahapan penelitian meliputi : (1) Pemurnian sampel dari ampul, (2) Pengujian nilai Minimum Inhibitory Concentration (MIC), (3) Membuat kurva pertumbuhan Pseudomonas aeruginosa D2 di berbagai konsentrasi CuSO4, (4) Menguji kemampuan akumulasi tembaga Pseudomonas aeruginosa D2 dengan Atomic Absorbtion Spectrophotometer (AAS), (5) Menguji kemampuan biosorpsi tembaga oleh Pseudomonas aeruginosa D2 dengan AAS. Hasil yang diperoleh menunjukan nilai MIC dari Pseudomonas aeruginosa D2 adalah 13 mM CuSO4. Bakteri tersebut dapat mengakumulasi 262,25 mg tembaga per gram berat kering sel pada konsentrasi 6 mM CuSO4 dan dapat mengabsorpsi 26,36 % CuSO4 pada media cair. / Waste from heavy metals has become one of the pollutant that requires serious attention because it is very dangerous. This is because heavy metals such as copper can affect the environment and living things easily. Copper itself can be used by living things as a micro-nutrient, one of which as cofactor, but in large amount it becomes toxic which can cause mutations. Solutions for remediation of copper in the environment have been carried out by chemical or physical means, but the method itself is less effective and has a relatively expensive operating costs. Therefore, a more economical and effective way is needed by using indigenous bacteria. Indigenous bacteria are bacteria isolated from places polluted by waste. In this study, an analysis of the accumulation and biosorption capacity of Pseudomonas aeruginosa D2 isolated from the Sukolilo River was carried out for copper. The stages of the research included: (1) Purification of samples from ampoules, (2) Testing the Minimum Inhibitory Concentration (MIC) value, (3) Making growth curves for Pseudomonas aeruginosa D2 at various concentrations of CuSO4, (4) Testing the ability of Pseudomonas aeruginosa D2 to accumulate copper with Atomic Absorption Spectrophotometer (AAS), (5) Testing the copper biosorption ability of Pseudomonas aeruginosa D2 with AAS. The results obtained showed that the MIC value of Pseudomonas aeruginosa D2 was 13 mM CuSO4. These bacteria can accumulate 262.25 mg per gram dry weight of cells at a concentration of 6 mM CuSO4 and can absorb 26.36% CuSO4 in liquid media.
