Abstract

Radikal bebas dapat disebabkan karena faktor intrinsik dan ekstrinsik. Faktor ekstrinsik seperti kondisi cuaca, pengaruh kondisi lingkungan, terpapar sinar UV, dan polusi dan dapat mengakibatkan kerusakan kulit, penuaan kulit terjadi lebih cepat, kering, munculnya flek hitam. Antioksidan merupakan suatu senyawa yang dapat meredam atau menangkal dampak negatif efek oksidan yang ada dalam tubuh dengan cara mendonorkan satu elektronnya kepada senyawa yang memiliki efek oksidan sehingga aktivitas senyawa oksidan dapat dihambat. Salah satu tanaman yang mempunyai aktivitas antioksidan adalah tanaman sarang semut (Myrmecodia tuberosa Jack) karena mempunyai kandungan senyawa flavonoid dan tanin. Penelitian bertujuan untuk menguji aktivitas antioksidan dari ekstrak etil asetat tanaman sarang semut (Myrmecodia tuberosa Jack) dengan menggunakan metode 1,1-Diphenyl-2-pycrylhydrazyl (DPPH). Sampel yang digunakan merupakan tanaman sarang semut (Myrmecodia tuberosa Jack) sebanyak 5 kg yang diperoleh dari Papua. Dalam pembuatan ekstrak dilakukan dengan metode maserasi menggunakan pelarut etil asetat. Pada penelitian, dilakukan skrining fitokimia untuk mengidentifikasi kandungan metabolit sekunder yang terdapat dalam suatu tanaman yang akan diteliti. Ekstrak etil asetat tanaman sarang semut (Myrmecodia tuberosa Jack) mempunyai kandungan senyawa metabolit sekunder seperti flavonoid, saponin, dan tanin. Pada penelitian diperoleh % rendemen sebesar 1,6932% dan kadar air sebesar 8,14%. Pada pengujian antioksidan menggunakan DPPH dengan parameter IC50 menunjukkan tanaman sarang semut (Myrmecodia tuberosa Jack) memiliki aktivitas antioksidan kategori kuat dengan IC50. Nilai IC50 ekstrak yang diperoleh sebesar 22,0394 μg/mL dan IC50 vitamin C sebesar 8,6990 μg/mL menunjukkan bahwa aktivitas antioksidan ekstrak lebih lemah dibandingkan antioksidan vitamin C. Aktivitas peredaman radikal bebas ekstrak etil asetat tanaman sarang semut (Myrmecodia tuberosa Jack) lebih lemah dibandingkan dengan vitamin C. / Free radicals can be caused by intrinsic and extrinsic factors. Extrinsic factors such as weather conditions, the influence of environmental conditions, exposure to UV rays, and pollution and can cause skin damage, skin aging occurs faster, dryness, the appearance of black spots. Antioxidants are compounds that can reduce or counteract the negative effects of oxidants in the body by donating an electron to compounds that have oxidant effects so that the activity of oxidant compounds can be inhibited. The aim of this study was to examine the antioxidant activity of the ethyl acetate extract of ant nests (Myrmecodia tuberosa Jack) using the 1,1- Diphenyl-2-pycrylhydrazyl (DPPH) method. The sample used was 5 kg of ant nests (Myrmecodia tuberosa Jack) obtained from Papua. Secondary metabolites found in ant nest plants (Myrmecodia tuberosa Jack) are flavonoids and tannins which have potential as antioxidants. In the manufacture of extracts carried out by maceration method using ethyl acetate solvent. In this study, phytochemical screening was carried out to identify the content of secondary metabolites present in a plant to be studied. Phytochemical screening conducted on the ethyl acetate extract of the ant nest plant (Myrmecodia tuberosa Jack) showed the presence of secondary metabolites such as flavonoids, saponins, and tannins. In this study, the % yield was 1.6932% and the water content was 8.14%. In the antioxidant test using DPPH with the IC50 parameter, the ant nest plant (Myrmecodia tuberosa Jack) has a strong category of antioxidant activity with IC50. The IC50 value of the extract obtained was 22.0394 μg/mL and the IC50 vitamin C was 8.6990 μg/mL indicating that the antioxidant power of the extract was weaker than the antioxidant vitamin C. The free radical scavenging activity of the ethyl acetate extract of the ant nest plant (Myrmecodia tuberosa Jack) weaker than vitamin C.
