Abstract

Tanaman sirih cina (Peperomia pellucida L. Kunth) dari keluarga piperaceae digunakan oleh masyarakat sebagai penyembuhan beberapa penyakit infeksi. Tumbuhan ini memiliki manfaat sebagai antibakteri pada pertumbuhan bakteri di area wajah seperti jerawat. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etil asetat sirih cina terhadap bakteri Propionibacterium acnes. Metode yang digunakan dalam penelitian adalah eksperimental laboratorium untuk pembuatan ekstrak etil asetat sirih cina, skrining fitokimia dan uji antibakteri. Metode yang digunakan pada pembuatan ekstrak adalah maserasi. Analisis data yang digunakan pada penelitian ini adalah SPSS tahun 2019 versi 26 dengan data yang terdistribusi normal karena p value > 0,05. Hasil yang diperoleh dari ekstrak etil asetat sirih cina memiliki % rendemen sebesar 11,53% dan % kadar air sebesar 2,02% serta mengandung senyawa metabolit sekunder seperti flavonoid, saponin dan steroid. Uji antibakteri menggunakan metode difusi sumuran dengan konsentrasi 750.000 ppm, 850.000 ppm dan 1.000.000 ppm dengan kontrol negatif aquadest dan kontrol positif vankomisin . Zona hambat yang terbentuk dari semua konsentrasi ekstrak etil asetat sirih cina masuk kedalam kategori CLSI yaitu “Intermediate” dan kontrol positif vankomisin masuk kedalam kategori “Susceptible”. Zona hambat yang dikategorikan efektif dalam menghambat pertumbuhan bakteri mempunyai nilai mean dengan rata-rata sebesar 18,877 pada konsentrasi 750.000 ppm. Dapat ditarik kesimpulan bahwa ekstrak etil asetat sirih cina mempunyai daya hambat dalam menghambat bakteri Propionibacterium acnes dengan kategori “Intermediate” / The sirih cina plant (Peperomia pellucida L. Kunth) from the piperaceae family is used by the community as a cure for several infectious diseases. This plant has the benefit of being an antibacterial against bacterial growth in areas of the face such as acne. This study aims to determine the antibacterial activity of sirih cina ethyl acetate extract against Propionibacterium acnes bacteria. The method used in this study was laboratory experimental for the manufacture of sirih cina ethyl acetate extract, phytochemical screening and antibacterial testing. The method used in the manufacture of extracts is maceration. Data analysis used in this study was SPSS year 2019 version 26 with normally distributed data because the p value > 0.05. The results obtained from the sirih cina ethyl acetate extract had a % yield of 11.53% and a % water content of 2.02% and contained secondary metabolites such as flavonoids, saponins and steroids. The antibacterial test used the well-diffusion method with concentrations of 750,000 ppm, 850,000 ppm and 1,000,000 ppm with aquadest negative control and vancomycin positive control. The inhibition zone formed from all concentrations of sirih cina ethyl acetate extract is included in the CLSI category, namely "Intermediate", and the vancomycin positive control is included in the "Susceptible" category. The inhibition zone which is categorized as effective in inhibiting bacterial growth has a mean value with an average of 18.877 at a concentration of 750,000 ppm. It can be concluded that the ethyl acetate extract of Chinese betel has an inhibitory power in inhibiting Propionibacterium acnes bacteria in the "Intermediate" category.
