Abstract

Methicillin-resistant Staphylococcus aureus menjadi salah satu strain dari bakteri Staphylococcus aureus yang mengalami resistensi terhadap antibiotik golongan beta-laktam dan antibiotik methicillin yang dapat diterapi menggunakan vankomisin. Tumbuhan genus Ficus menjadi salah satu pengobatan alternatif bahan alam yang dimanfaatkan sebagai antibakteri dengan metabolit sekunder flavonoid, tritepenoid, dan steroid. Penelitian ini ditujukan untuk mengetahui Konsentrasi Hambat Minimum dan Konsentrasi Bunuh Minimum ekstrak n-heksan, ekstrak etil asetat, dan ekstrak etanol 96% dari daun Ficus virens glabella Warb maupun daun Ficus callosa Warb pada pertumbuhan bakteri MRSA. Ekstrak n-heksan, ekstrak etil asetat, dan ekstrak etanol 96% dibuat dengan ekstraksi bertingkat reflux. Penentuan nilai KHM dan KBM menggunakan metode mikrodilusi. Hasil penelitian menunjukkan kedua tanaman diduga memiliki kemampuan penghambatan terhadap bakteri MRSA yang ditunjukkan dengan konsentrasi 10.000 ppm kedua ekstrak sampel daun memiliki nilai persen penghambatan yang mendekati nilai persen penghambatan vankomisin, namun belum dapat disimpulkan nilai KHM secara lebih lanjut karena hasil dari analisis statistik menunjukkan tidak adanya perbedaan bermakna dari kelompok pelarut yang digunakan terhadap nilai persen penghambatan pertumbuhan bakteri. Nilai KBM belum dapat disimpulkan mengingat penghambatan telah mencapai konsentrasi tertinggi. / Methicillin-resistant Staphylococcus aureus is one strain of Staphylococcus aureus bacteria that has resistance to beta-lactam antibiotics and methicillin antibiotics which can be treated using vancomycin. Ficus genus plants are one of the alternative natural treatments that are utilized as antibacterials with secondary metabolites of flavonoids, tritepenoids, and steroids. This study aims to determine the Minimum Inhibitory Concentration and Minimum Bactericidal Concentration of n-hexane extract, ethyl acetate extract, and 96% ethanol extract from Ficus virens glabella Warb leaves and Ficus callosa Warb leaves on MRSA bacterial growth. The n-hexane extract, ethyl acetate extract, and 96% ethanol extract were prepared by reflux multistage extraction. Determination of MIC and MBC values using microdilution method. The results showed that both plants were suspected of having inhibitory ability against MRSA bacteria as indicated by the concentration of 10,000 ppm, both leaf sample extracts had a percent inhibition value that was close to the percent inhibition value of vancomycin, but the MIC value could not be concluded further because the results of statistical analysis showed no significant difference from the solvent group used on the percent inhibition value of bacterial growth. The MBC value cannot be concluded considering the inhibition has reached the highest concentration.
