Abstract

Berenuk (Crescentia cujete L.) adalah tanaman dari suku Bignoniaceous. Tanaman ini dapat tumbuh di daerah asia. Tanaman ini banyak di manfaatkan oleh masyarakat Filipina sebagai obat tradisional namun belum terlalu banyak masyarakat Indonesia yang mengetahui khasiat dari tanaman ini. Pada penelitian ini, ekstrak buah berenuk (C.cujete L.) di uji pada mencit untuk mengetahui efektivitas antidiare. Tujuan dilakukannya penelitian ini untuk mengetahui kandungan metabolit sekunder buah berenuk sebagai antidiare dan mengetahui efektivitas ekstrak buah berenuk sebagai antidiare pada mencit. Penelitian ini dilakukan ekstraksi menggunakan metode maserasi bertingkat dengan pelarut etanol 70%. Hasil skrining fitokimia diperoleh bahwa buah berenuk mengandung senyawa Flavonoid, Alkaloid, Tanin, Fenol dan Saponin. Uji KLT di hasilkan ekstrak buah berenuk (C.cujete L.) mengandung senyawa kuersetin yang merupakan senyawa turunan dari flavonoid. Selanjutnya untuk melihat aktivitas antidiare dilakukan pengujian dengan metode transit intestinal. Parameter uji dari metode transit intestinal adalah melihat perbandingan antara panjang marker yang melalui usus dengan panjang keseluruhan usus. Nilai rasio usus dari pengujian ekstrak buah berenuk pada mencit yaitu kontrol negatif (0,61± 0.078), kontrol positif (0,28±0.028) dan kelompok ekstrak 300mg/kg BB (0,48±0.035), 600mg/kgBB (0,38±0.039) dan 1200mg/kgBB (0,33±0.017). Data kemudian dianalisis secara statistik dengan metode kruskal wallis dan dunn test. Hasil yang diperoleh adalah kelompok kontrol positif dengan kelompok ekstrak buah berenuk (C.cujete) 1200mg/kg tidak berbeda signifikan sehingga dari pengujian ini, ekstrak buah berenuk (C.cujete) 1200mg/kg merupakan dosis yang efektif sebagai antidiare. / Calabash (Crescentia cujete L.) is a plant from the Bignoniaceous tribe. This plant can grow in Asia. This plant is widely used by Filipino people as traditional medicine, but not too many Indonesian people know the benefits of this plant. In this study, calabash fruit extract (C.cujete L.) was tested on mice to determine its antidiarrheal effectiveness. The aim of this research was to determine the secondary metabolite content of berenuk fruit as an anti-diarrhea and to determine the effectiveness of berenuk fruit extract as an anti-diarrhea in mice. This research carried out extraction using a multilevel maceration method with 70% ethanol solvent. The results of phytochemical screening showed that calabsh fruit contains flavonoids, alkaloids, tannins, phenols and saponins. The TLC test resulted in calabash fruit extract (C. cujete L.) containing the compound quercetin. After that, to know the antidiarrheal activity, testing was carried out using the intestinal transit method. The test parameter of the intestinal transit method is to look at the comparison between the length of the marker that passes through the intestine and the length of the entire intestine. The intestinal ratio values from testing calabash fruit extract in mice were negative control (0.61±0.078), positive control (0.28±0.028) and extract groups 300mg/kg BW (0.48), 600mg/kgBW (0.38±0.039) and 1200mg/ kgBB (0.33±0.017). The data was then analyzed statistically using the Kruskal Wallis and Dunn test. The results is the positive control group with the 1200 mg/kg calabsh (C.cujete L.) fruit extract group which was not significantly differen, calabash fruit extract (C.cujete L.) 1200mg/kg is an effective dose as an antidiarrheal.
