Abstract

Diabetes adalah masalah kesehatan global yang diderita jutaan orang dan membutuhkan pengobatan yang efektif. Indonesia dianugerahi kekayaan sumber daya alam, salah satunya adalah tanaman obat. Dalam penelitian ini dilakukan eksplorasi potensi antidiabetes dari ekstrak Castanopsis tungurrut dengan menggunakan metode in vitro dan in silico. Pengujian in vitro melibatkan eksperimen langsung pada proses seluler terkait dengan diabetes dengan fokus khusus pada aktivitas penghambatan enzim α-amilase dan α-glukosidase. Sementara itu, metode in silico diterapkan untuk menganalisis interaksi molekuler antara senyawa dan reseptor yang terkait dengan penghambatan enzim tersebut. Acarbose sebagai obat yang bekerja dengan mekanisme yang sama digunakan sebagai pembanding. Pengujian in vitro menunjukkan bahwa ekstrak batang Castanopsis tungurrut menghasilkan aktivitas penghambatan dengan nilai IC50 sebesar 156,524 ppm pada α-amilase dan 3,449 ppm untuk α-glukosidase dengan mode penghambatan secara kompetitif. Hasil uji in silico mengidentifikasi senyawa potensial sebagai kandidat obat antidiabetes, yaitu casuarinin yang memiliki nilai afinitas lebih baik daripada acarbose pada kedua enzim tersebut. / Diabetes is a global health problem that affects millions of people and requires effective treatment. Indonesia is blessed with a wealth of natural resources, one of which is medicinal plants. In this study, the antidiabetic potential of Castanopsis tungurrut extract was explored using in vitro and in silico methods. In vitro testing involves direct experimentation on cellular processes related to diabetes with a particular focus on the inhibitory activity of α-amylase and α-glucosidase enzymes. Meanwhile, the in silico method was applied to analyze the molecular interactions between the compound and the receptors associated with the inhibition of these enzymes. Acarbose as a drug that works with the same mechanism was used as a comparator. Results showed that Castanopsis tungurrut stem extract produced inhibitory activity with IC50 values of 156,524 ppm on α-amylase and 3,449 ppm for α-glucosidase with a competitively inhibitory mode. In silico test results identified potential compounds as antidiabetic drug candidates, namely casuarinin which has better affinity value than acarbose on both enzymes.
