Abstract

Bakteriosin merupakan peptida yang disintesis secara ribosomal oleh bakteri asam laktat (BAL). Produksi bakteriosin oleh BAL sangat dipengaruhi oleh komposisi media kultur yang harus mengandung sumber nitrogen, karbohidrat, asam amino, protein, vitamin, serta mineral. Oleh karena itu, sintesis bakteriosin dapat ditingkatkan melalui optimasi media. Salah satu media yang dapat digunakan untuk pertumbuhan bakteri asam laktat adalah whey tahu. Whey tahu kaya akan nutrisi penting sehingga diharapkan dapat mendukung pertumbuhan BAL serta produksi bakteriosin. Lactiplantibacillus plantarum SU-KC1a koleksi Laboratorium Biologi Universitas Pelita Harapan diketahui memiliki gen pengode bakteriosin (plantaricin). Selain itu, terdapat pula koleksi BAL lain yang diisolasi dari duodenum babi yang sehat, yaitu Weisella confusa D1. Oleh karena itu, pada penelitian ini potensi whey tahu sebagai media pertumbuhan BAL dan produksi bakteriosin oleh L. plantarum SU-KC1a dan W. confusa D1 akan diteliti. Hasil penelitian menunjukkan L. plantarum SU-KC1a dan W.confusa D1 dapat tumbuh pada media whey tahu. Selain itu, terdapat aktivitas antibakteri dari co-culture L. plantarum SU-KC1a dan W. confusa D1. Aktivitas antibakteri paling baik berada pada L. plantarum SU-KC1a (10^7 CFU/mL) dan W. confusa D1 (10^6 CFU/mL) pada waktu panen 42 jam dengan penambahan L. plantarum SU-KC1a jam ke 3, 6, dan 9 pada kultur W. confusa D1. Berat protein belum dapat diidentifikasi melalui Tricine SDS Page dikarenakan kemurnian protein rendah, sehingga perlu metode tambahan terkait kemurnian protein. / Bacteriocins are peptides synthesized ribosomally by lactic acid bacteria (LAB). Bacteriocin production by LAB is strongly influenced by the composition of the culture medium, which must contain sources of nitrogen, carbohydrates, amino acids, proteins, vitamins, and minerals. Therefore, bacteriocin synthesis can be improved through media optimization. One of the media that can be used for the growth of lactic acid bacteria is tofu whey. Tofu whey is rich in essential nutrients that are expected to support LAB growth and bacteriocin production. Lactiplantibacillus plantarum SU-KC1a from the Biology Laboratory of Universitas Pelita Harapan is known to have a bacteriocin (plantaricin) encoding gene. In addition, there is also another LAB collection isolated from the duodenum of healthy pigs, namely Weisella confusa D1. Therefore, in this study, the potential of tofu whey as a LAB growth medium and bacteriocin production by L. plantarum SU-KC1a and W. confusa D1 will be investigated. The results showed that L. plantarum SU-KC1a and W. confusa D1 could grow on tofu whey media. In addition, there was antibacterial activity from the co-culture of L. plantarum SU-KC1a and W. confusa D1. The best antibacterial activity was in L. plantarum SU-KC1a (10^7 CFU/mL) and W. confusa D1 (10^6 CFU/mL) at harvest time 42 hours with the addition of L. plantarum SU-KC1a at 3, 6, and 9 hours in W. confusa D1 culture. Protein weight cannot be identified through Tricine SDS Page due to low protein purity, so additional methods related to protein purity are needed.
