Abstract

Lebah madu merupakan polinator alami yang berperan besar dalam proses penyerbukan tanaman agrikultur. Namun, saat ini terjadi penurunan populasi lebah madu yang disebabkan oleh beberapa faktor, seperti perubahan cuaca, konversi lahan, dan infeksi patogen. Penurunan populasi lebah madu akan berdampak pada penurunan produksi tanaman agrikultur dan ketahanan pangan. Maka dari itu, diperlukan penelitian mengenai komunitas bakteri pada usus lebah madu, yang merupakan faktor utama dalam memengaruhi kesehatan lebah madu itu sendiri. Isolasi DNA genom dan amplifikasi 16S rDNA komunitas bakteri dalam midgut dan hindgut lebah madu A. cerana dilakukan terlebih dahulu lalu dilanjutkan dengan analisis metagenomik. Analisis metagenomik merupakan pendekatan molekuler yang dapat menunjukkan keberagaman dan kelimpahan komunitas bakteri, menganalisis anotasi spesies dan konstruksi hubungan filogenetik, serta menghasilkan alpha diversity analysis dalam suatu sampel dengan memanfaatkan berbagai program bioinformatika. Pada penelitian ini digunakan Ion S5XL untuk proses sequencing analisis metagenomik. Hasil penyaringan 140.345 data Clean Reads didapatkan 103.931 data Taxon tag lalu dikelompokkan menjadi 63 OTU. Berdasarkan alpha diversity analysis, 63 OTU yang dihasilkan tersebut telah dapat mengilustrasikan keseluruhan komunitas bakteri dalam usus lebah madu A. cerana. Sepuluh genus dengan kelimpahan relatif terbanyak yang didapatkan dari analisis metagenomik usus lebah madu A. cerana, yaitu Gilliamella, Lactobacillus, Snodgrassella, Apibacter, Bifidobacterium, Klebsiella, Serratia, Stenotrophomonas, Acinetobacter, dan Commensalibacter. Kelimpahan relatif Gilliamella yang lebih tinggi dibandingkan kelimpahan relatif Lactobacillus, Bifidobacterium, Commensalibacter, dan Snodgrasella menandakan bahwa koloni lebah madu A. cerana yang digunakan dalam penelitian ini memiliki mikrobiota yang tidak seimbang. / Honey bees are natural pollinators that play a major role in the process of agricultural crops pollination. However, currently there is a decline in honey bee population which is caused by several factors, such as climate changes, land conversion, and pathogen infections. The decline in honey bee population could lead to a major decline in agricultural crop production which eventually affecting food security. Therefore, research on the bacterial community of the honey bee intestine, which is a major factor in influencing the health of the honey bee itself, is needed. Isolation of genomic DNA and 16S rDNA amplification of the bacterial community in the midgut and hindgut of honey bee A. cerana was carried out first then continued with metagenomic analysis. Metagenomic analysis is a molecular approach that can show diversity and abundance of bacterial communities, analyze species annotations, construct phylogenetic relationships, and produce alpha diversity analysis in a sample by utilizing various bioinformatics programs. In this study Ion S5XL was used for the sequencing process of metagenomic analysis. The results of the 140,345 Clean Reads data obtained 103,931 Taxon tag data which then grouped into 63 OTUs. Based on alpha diversity analysis, the 63 OTUs that have been produced can illustrate the entire bacterial community in the intestines of A. cerana honey bees. Ten genera with the most relative abundance in A. cerana honey bee gut were Gilliamella, Lactobacillus, Snodgrassella, Apibacter, Bifidobacterium, Klebsiella, Serratia, Stenotrophomonas, Acinetobacter, and Commensalibacter. The relative abundance of Gilliamella which is higher than the relative abundance of Lactobacillus, Bifidobacterium, Commensalibacter, and Snodgrasella indicates that the honey bee colony of A. cerana used in this study has an unbalanced microbiota.
