Abstract
infeksi patogen dengan memproduksi komponen antibakteri seperti asam laktat dan bakteriosin. Pada penelitian ini, aktivitas antibakteri dari L. plantarum F75 dievaluasi dengan well diffusion assay, broth dilution assay dan time-kill test (co-culture). Berdasarakan hasil yang diperoleh dari well diffusion assay, cell-free supernatant (CFS) L. plantarum F75 menunjukkan aktivitas antibakteri terhadap E. coli dan S. aureus. Efek penghambatan tersebut dipengaruhi oleh nilai pH, penghambatan yang lebih baik ditunjukkan pada nilai pH yang lebih rendah. Perlakuan CFS pada suhu tinggi (100-121°C) mengurangi efek penghambatan terhadap pertumbuhan S. aureus, namun tidak ada penurunan efek penghambatan terhadap pertumbuhan E. coli. Hasil dari broth dilution assay menunjukkan bahwa persentase minimal CFS untuk menghambat kedua patogen (MIP) adalah 10%, sedangkan persentase minimal CFS untuk membunuh (MBP) E. coli dan S. aureus masing-masing adalah 20% dan 40%. Pengujian time-kill test menunjukkan bahwa berbagai jumlah sel L. plantarum F75 (105, 106 dan 107 CFU/ml) mampu membunuh patogen (dalam kultur campuran) secara total setelah 24 jam inkubasi./ Lactobacillus plantarum is one of the probiotics that has the potential to fight pathogenic infections by producing antibacterial components such as lactic acid and bacteriocin. In this study, antibacterial activity of L. plantarum F75 was evaluated with well diffusion assay, broth dilution assay and time-kill test (co-culture). Based on the results obtained from well diffusion assay, cell-free supernatant (CFS) of L. plantarum F75 showed antibacterial activity against E. coli and S. aureus. The inhibitory effect was influenced by pH values, better inhibition was shown in lower pH values. CFS treatment at high temperatures (100-121°C) reduced the inhibitory effect on the growth of S. aureus growth, but not on E. coli growth. Results from broth dilution assay showed that the minimum percentage of CFS to inhibit both pathogens (MIP) was 10%, while the minimum percentage of CFS to kill (MBP) E. coli and S. aureus was 20% and 40% respectively. Time-kill testing showed that various number of L. plantarum F75 cells (105, 106 and 107 CFU/ml) were able to kill both pathogens cells (in mixed cultures) after 24 hours of incubation.
