Abstract

Terdapat beberapa produk komeril seperti Starter Culture Yoghurt dan suplemen yang mengandung bakteri probiotik. Salah satu bakteri yang sering dipakai dalam produk ini adalah Bifidobacterium sp. Penelitian ini bertujuan untuk mengisolasi, mengidentifikasi, serta mengkarakterisasi bakteri anaerobik obligat Bifidobacterium sp. dari Starter culture Yoghurt dan suplemen yang sejauh ini belum dilakukan di Indonesia. Pada penelitian ini sampel diperoleh dari starter culture Chris Hansen dan suplemen BION 3. Metode yang dilakukan diantaranya adalah menumbuhkan bakteri dari kedua sampel pada media MRS (De Mann, Rogosa, dan Sharpe) dan TPY (Trypticase Phytone Yeast Extract) yang dimodifikasi menjadi media selektif untuk Bifidobacteria; identifikasi morfologi; dan karakterisasi isolat (uji aktivitas enzim katalase dan uji motilitas agar); uji ketahanan pH dan garam empedu; serta uji suseptibilitas terhadap berbagai antibiotik. Untuk mengetahui spesies Bifidobacterium yang didapat dilakukan sequencing 16S. Jenis media terbaik untuk pertumbuhan bakteri dari sampel yang diperoleh adalah media MRS (De Mann, Rogosa, dan Sharpe) agar dan broth dengan modifikasi penambahan L-cysteine dan mupirocin. Isolat CHSC dan STBM diperoleh dari sampel starter culture dan suplemen dengan karakterisik grampositif, non-motil, dan katalase negatif. Dari hasil evaluasi potensi probiotik, isolat CHSC dan STBM berpotensi sebagai probiotikBerdasarkan hasil sequencing 16srRNA, isolat CHSC diketahui merupakan spesies Bifidobacterium animalis subsp. lactis. Isolat STBM memiliki kemungkinan sebagai spesies yang sama dengan isolat CHSC atau Bifidobacterium longnum. Penelitian lebih lanjut perlu dilakukan untuk memastikan spesies dari isolat STBM. / There are various commercial products that contain probiotic bacteria such as yoghurt and supplement. One of the probiotic bacteria that is commonly used in these commercial products is Bifidobacterium sp. This research aims to isolate, identify, and characterized the anaerobic obligate bacteria Bifidobacterium sp. from yoghurt starter culture and supplement that has not been done in Indonesia. In this research, Chris Hansen starter culture and Bion 3 supplement is used as samples. Two types of selective growth media (modified De Mann, Rogosa, dan Sharpe and Trypticase Phytone Yeast Extract agar) is evaluated in this research to determine the most suitable for Bifidobacterium.Morphology and characteristic of the isolates obtained is also identified using gram staining, catalase test, and motility test. Isolates potential as probiotic is also evaluated using acidity and bile salt tolerance and antibiotic susceptibility. 16S RNA sequencing is used to identify the species of the isolates.MRS agar and broth with addition of L-cysteine and mupirocin is found to be the most suitable media for the isolates. Isolates CHSC and STBM is obtained from starter culture and supplement respectively with the characteristic of gram positive, nonmotile, and catalase negative for both isolates. Results showed CHSC and STBM exhibit probiotic potential, although further research is required to support this claim. 16S sequencing show that CHSC is Bifidobacterium animalis subsp. lactis, while STBM is still undecide between the same species with CHSC or Bifidobactreium longnum. Further research is needed to decide the species of isolate STBM.
