Abstract
Luka merupakan salah satu jalur yang dapat digunakan oleh bakteri patogen oportunistik untuk menginfeksi tubuh. Jaringan yang terbuka akibat luka merusak barrier pertahanan pertama tubuh dan membuatnyarentan terhadap infeksi dari patogen oportunis. Sebuah studi, tingkat kematian yang disebabkan oleh infeksi akut berkisar antara 40% hingga 58%. Pseudomonas aeruginosa merupakan salah satu bakteri yang menyebabkan insiden infeksi yang tinggi. Sebuah studi mengungkapkan bahwa tingkat insidien infeksi oleh P. aeruginosa mencapai 89% dari total kasus infeksi yang terjadi. P. aeruginosa juga dilaporkan memiliki resistensi terhadap berbagai antibiotis dan antiseptik, dan dinyatakan sebagai bakteri multi-drug resistant (MDR). Salah satu alternatif yang sedang dikembangkan adalah penggunaan probiotik untuk mencegah infeksi dan mempercepat penyembuhan luka. Oleh sebab itu, tujuan dari penelitian ini adalah menyelidiki efek dari pemberian probiotik Lactobacillus fermentum strain E5 secara oral dan topikal pada penyembuhan luka pada mencit Balb/c yang diinfeksikandengan Pseudomonas aeruginosa. Parameter yang diuji adalah uji inhibisi in vitro L. fermentum strain E5, laju penutupan luka, jumlah CFU bakteri di sekitar jaringan luka,dan tingkat kematian. Hasil menunjukkan bahwa perlakuan dengan probiotik dapat secara signifikan mempercepat laju penutupan luka, mengurangi resiko kematian, dan resiko infeksi luka. Mencit yang diberikan probiotik dengan metode topikal menunjukkan laju penutupan paling cepat dan memiliki tingkat kematian terendah. / Wound are one of many pathways that can serve as an entry point for pathogen to infect its host’s body. Wounded tissue will cause the skin barrier to tear open, making it vulnerable to infection from opportunistic pathogen. Death rates that were caused by acute infection ranged from 40% to 58%, and Pseudomonas aeruginosa is one of the leading causes for acute wound infection. Studies showed that P. aeruginosa infection has an incidence rate up to 89% from all infection cases. Recent papers reported that P. aeruginosa has expressed resistance to various antibiotics and antiseptics, and has been labeled as Multi- Drug Resistant (MDR) pathogen. One of the alternatives that are being developed is the utilization of probiotics for treating wounds. That being said, the goal of this research is to determine the effect of utilizing Lactobacillus fermentum strain E5, from UPH bacteria collection, for treating P. aeruginosa infected wounds on Balb/C mice. Parameters that were measured were wound healing rate, bacteria cell count on the wounded tissue, mortality rate, and in-vitro inhibition activity of L. fermentum strain E5. Results showed mice that were treated with probiotics had a significant increase in wound healing rate, lower mortality rate, and lower infection risk, compared to control groups. Mice that were treated with probiotic topically showed faster wound healing rate and had the lowest mortality rate.
