Abstract
Limbah industri tekstil merupakan salah satu masalah pencemaran lingkungan yang terjadi di Indonesia. Pewarna tekstil yang terdapat pada limbah industri tekstil menjadi polutan yang dapat bersifat toksik bagi lingkungan. Salah satu pewarna tekstil yang digunakan adalah pewarna methylene blue (MB). Methylene blue merupakan pewarna kationik yang umum digunakan dalam industri tekstil, kertas, plastik, maupun kulit. Limbah industri tekstil yang mengandung pewarna sayangnya seringkali dibuang secara langsung ke perairan sehingga menyebabkan pencemaran lingkungan. Hal ini juga terjadi pada Sewon dan Kulon Progo yang terdapat di Daerah Istimewa Yogyakarta yang memiliki industri tekstil yang membuang limbah tekstil tanpa diolah terlebih dahulu. Bioremediasi menjadi solusi yang efektif untuk mendegradasi pewarna MB. Bakteri yang diperoleh dari DIY diisolasi, dikarakterisasi dan diidentifikasi. Isolat bakteri yang diperoleh adalah Comamonas aquatica dan Ralstonia pickettii yang diidentifikasi dengan metode DNA sequencing 16S rRNA. C. aquatica dan R. pickettii berturut-turut dapat mendekolorisasi pewarna MB hingga 78,4% dan 71,7% pada suhu 37 oC selama 96 jam pada media glucose yeast peptone (GYP) yang ditambahkan dengan pewarna MB 0,005% (w/v). / Effluent from textile industry is a common problem that has happened in Indonesia. Textile dye that has a deleterious effect is released to the environment without going through any wastewater treatment. One of the most used dye for textile industry is methylene blue. Methylene blue is a common dye for textile industry, paper, plastic and leather. Effluent from textile industry usually does not undergo any wastewater treatment, this phenomenon is also happening in Sewon and Kulon Progo at Yogyakarta which has many textile industries that do not pay attention to this problem. Bioremediation is an effective solution to degrade methylene blue. Bacteria from Yogyakarta sewage was isolated, characterized and identified. These isolates of bacteria are Comamonas aquatica and Ralstonia pickettii. Bacteria was identified using 16S rRNA DNA sequencing method. C.aquatica and Ralstonia pickettii have the ability to decolorize bacteria until 78,4% and 71,7% decolorization respectively on glucose yeast peptone medium with addition of 0,005% (w/v) methylene blue
