Abstract

Tingginya ekspor udang windu (P. monodon) di Indonesia menyisakan limbah kepala dan kulit udang yang dapat diolah lebih lanjut menghasilkan glukosamin, namun depolimerisasi secara kimiawi menimbulkan banyak dampak negatif bagi lingkungan sehingga hidrolisis kitin menggunakan bakteri menjadi alternatif utama. Penelitian ini bertujuan mengetahui bakteri yang memiliki daya kitinolitik kuat dari kulit udang windu. Penelitian dimulai dengan preparasi kitin sebagai substrat dalam bentuk kitin koloidal pada media isolasi, pemurnian, dan uji daya bakteri kitinolitik. Karakterisasi morfologi koloni dan sel dilakukan untuk dapat membedakan isolat yang diperoleh selama tahap pemurnian. Daya kitinolitik bakteri diuji berdasarkan analisa parameter indeks kitinolitik dan pengukuran parameter warna serta analisis tingkat lightness media daya kitinolitik yang ditambahkan pewarna indikator Bromocresol Purple (BCP). Bakteri terpilih dengan daya kitinolitik paling kuat diidentifikasi molekuler berdasarkan sekuens 16S rDNA. Isolat yang berhasil diperoleh dari kulit udang windu adalah berjumlah 17 isolat dengan 8 isolat bakteri memiliki daya kitinolitik kuat. Isolat A12 terpilih sebagai isolat bakteri dengan daya kitinolitik terkuat berdasarkan nilai indeks kitinolitik terbaik, yakni 4,46 dan parameter lightness terendah, yakni 23,88. Isolat A12 diidentifikasi merupakan Providencia stuartii dengan kemiripan 99,33%. / High export of Black Tiger Prawn (P. monodon) in Indonesia leaves high shrimp shell waste which can be further processed to produce glucosamine, but chemical depolymerization causes many negative impacts to the environment so that chitin hydrolysis using bacteria becomes the main alternative. The purpose of this study was to determine the bacteria which have strong chitinolytic activity from Black Tiger Prawn exoskeleton. The study began with chitin preparation which will be used as substrate in the form of colloidal chitin in isolation, purification, and chitinolytic assay media. Characterization of colony and cell morphology was performed to distinguish isolates obtained during the purification step. Chitinolytic activity of bacteria was analyzed based on parameters of Chitinolytic Index and the color lightness measurement of media with addition of Bromocresol Purple (BCP) dye indicator. The preferred bacteria with the strongest chitinolytic activity were identified molecularly based on the 16S rDNA sequence. The isolates obtained from Black Tiger Prawn shell were 17 bacterial isolates with 8 strong chitinolytic isolates. Isolate A12 was selected as the strongest chitinolytic bacteria based on its best IK value (4.46) and darkest lightness parameter (23.88). Isolate A12 was identified as Providencia stuartii with 99.33% similarity.
