Abstract

Henna (Lawsonia inermis L.) is a type of plant that has been used as a source of natural dye for skin and hair since 1400 BC. The ability of this plant as a coloring compound is due to the presence of natural pigment called lawsone (2-hydroxy-1,4- napthoquinone). Lawsone is a major bioactive component that is known to have various biological activities such as bactericidal, fungicidal, antimalarial, cytostatic, and antioxidant properties. Lawsone is an interesting compound to scientists because of its ability to form its derivative compounds, which are known to have an increase in therapeutic effects. This research was conducted to identify the presence of lawsone in the leaves and to modify the structure of lawsone itself. The identification process was first carried out by macerating the leaves using three types of solvents with different polarity, namely n-hexane, ethyl acetate, and ethanol. The obtained crude extracts were tested to determine the yield percentage, total phenolic content, antioxidant activity, Rf value, maximum wavelength, and mass spectrum (GC-MS) to confirm the existence of lawsone in the crude extract of Henna leaves. The results of the identification showed that lawsone can be obtained by using ethyl acetate in the maceration process. The obtained crude extract from ethyl acetate was chosen to be proceed in the research stage II. In research stage II, the study was carried out by adding ethanolamine (0.15 M) to the chosen crude extract (0.1 M) in different solvent conditions (water, ethanol) and different reaction times (15 min., 30 min., 45 min.). The products were analyzed to find out the yield percentage, Rf value, maximum wavelength, IC50 value, and mass spectrum of the formed compound. The expected product with m/z of 233 was found through GC-MS analysis. Based on statistical analysis, there was a significant difference (p < 0.05) between the original crude extract and after modification, with an increase in antioxidant activity for 8.4940%.
