Abstract

Reactive black B (RBB) adalah salah satu jenis pewarna azo yang umum digunakan. Pewarna RBB diketahui bersifat mutagenik dan karsinogenik. Sifat mutagenik tersebut berperan dalam aktivitas mutagenik yang terdapat pada daratan atau permukaan air yang tercemar oleh limbah tekstil. Cara untuk menanggulangi limbah tekstil sudah banyak dikembangkan, namun belum banyak diketahui sifat toksisitas dari hasil degradasi limbah pewarna tersebut. Penelitian ini dilakukan untuk mengevaluasi kemampuan bakteri koleksi UPH dalam mendegradasi dan menganalisis metabolit yang dihasilkan. Berdasarkan evaluasi kemampuan dekolorisasi pada medium cair, terdapat dua isolat unggul dari total keseluruhan tujuh isolat yang diuji. Kedua isolat tersebut adalah isolat RBB 2 dan KRBB 4.1 dengan kemampuan dekolorisasi tertinggi 45,81% dan 55,80 % secara berurutan. Pada uji toksisitas, metabolit yang dihasilkan tidak bersifat toksik terhadap sel bakteri maupun tumbuhan kacang hijau dan padi. Sedangkan berdasarkan karakterisasi enzim yang dilakukan dengan SDS-PAGE dapat diketahui bahwa enzim yang berperan adalah NADH-flavin azoreduktase (85 kDa) dan lakase (63 kDa).
