Abstract
Pada penelitian kali ini, Lactobacillus plantarum F75 dan Lactobacillus casei 97/L3 dimikroenkapsulasi menggunakan natrium alginat (4%) serta kitosan (0,75%) melalui teknik ekstrusi. Mikrokapsul yang terbentuk kemudian di freeze dry untuk mengurangi konten air yang terkandung dan memperpanjang shelf-life. Suspensi sel dan mikrokapsul disimpan pada suhu kulkas (4°C) serta suhu ruang selama 28 hari. Setelah 28 hari, sel dan mikrokapsul yang menunjukkan stabilitas tinggi adalah yang disimpan pada suhu 4°C. Uji in vitro juga dilakukan pada simulated gastric juice (pH 2,0) dan intestinal juice (garam empedu) selama 120 menit pada suhu 37°C untuk mengetahui tingkat keselamatan probiotik bebas maupun yang dimikroenkapsulasi. Hasilnya menunjukkan bahwa mikroenkapsulasi menggunakan natrium alginat – kitosan sukses melindungi probiotik secara signifikan pada pH yang rendah, namun tidak pada lingkungan yang mengandung garam empedu
