Abstract
Escherichia coli dan Staphylococcus aureus merupakan bakteri patogen oportunistik yang dapat menyebabkan infeksi gastrointestinal. Probiotik dapat digunakan sebagai agen preventif untuk menghambat pertumbuhan patogen yang berlebihan pada saluran pencernaan. Pada penelitian ini, dilakukan evaluasi penghambatan (antibakteri) dari Lactobacillus plantarum F75 terhadap
pertumbuhan E. coli dan S. aureus. Aktivitas antibakteri dari L. plantarum F75 dievaluasi dengan metode well diffusion, time-kill test (co-culture) dan broth macrodilution assay, dengan menggunakan cell free supernatant (CFS). Hasil penelitian menunjukkan bahwa L. plantarum F75 dengan berbagai konsentrasi mampu menghambat pertumbuhan E. coli dan S. aureus. Kematian sel patogen sebesar 100% terjadi pada rentang waktu 10-16 jam setelah dikultur bersama L. plantarum F75. Persentase minimum CFS yang dibutuhkan untuk menghambat (MIP) E. coli dan S. aureus adalah masing-masing 10-20% dan 10%, sedangkan persentase minimum CFS untuk membunuh (MBP) kedua patogen secara berturutturut adalah 40% dan 30%. Nilai MIP dan MBP menunjukkan bahwa S. aureus lebih rentan terhadap CFS L. plantarum F75 dibandingkan E. coli. Time-kill test dan broth macrodilution assay mengindikasikan bahwa asam laktat yang dihasilkan L. plantarum F75 mengakibatkan penurunan pH pada medium kultur sehingga menyebabkan penghambatan pertumbuhan E. coli dan S. aureus. Selain menghasilkan asam L. plantarum F75 diduga memproduksi komponen antibakteri lainnya, seprti bakteriosin. / Escherichia coli and Staphylococcus aureus are opportunistic pathogens that can cause gastrointestinal infections. Probiotics can be used as preventive agents to inhibit the overgrowth of pathogens in the digestive tract. This study evaluated the inhibition (antibacterial) of Lactobacillus plantarum F75 against the growth of E. coli and S. aureus. The antibacterial activity of L. plantarum F75 was evaluated
using the well diffusion method, time-kill test (co-culture), and broth macrodilution assay, using cell-free supernatant (CFS). The results showed that L. plantarum F75 with various concentrations was able to inhibit the growth of E. coli and S. aureus. 100% of pathogenic cell deaths occurred in 10-16 hours after being cultured with
L. plantarum F75. The minimum percentage of CFS required to inhibit (MIP) E. coli and S. aureus is 10-20% and 10%, respectively, while the minimum percentage of CFS to kill (MBP) of the two pathogens is 40% and 30%, respectively. The MIP and MBP values indicated that S. aureus was more susceptible to CFS L. plantarum F75 than E. coli. Time-kill test and broth macrodilution assay indicated that the lactic acid produced by L. plantarum F75 resulted in a decrease in pH in the culture medium which resulted in inhibition of the growth of E. coli and S. aureus. In addition to producing L. plantarum F75 acid, it is thought to produce other antibacterial components, such as bacteriocins.
