Abstract

Limbah kulit udang windu (Penaeus monodon) mengandung kitin yang sifatnya sukar larut karena berikatan dengan senyawa lain seperti protein dan asam amino, namun memiliki banyak manfaat dalam bidang makanan, agrikultural, obat-obatan dan bangunan, sehingga kitin harus dibuat produk turunannya terlebih dahulu agar pemanfaatnnya optimal. Tujuan dari penelitian ini adalah untuk mengisolasi dan mengidentifikasi kapang yang memiliki daya kitinolitik kuat yang diisolasi dari kulit udang windu. Tepung kulit udang diperoleh dengan pengeringan kulit udang dengan matahari dan pengecilan ukuran, lalu dibusukkan dan dilakukan pour plate pada tiga tingkat pengenceran 10-3, 10-4, dan 10-5, kemudian dilakukan isolasi dan pemurnian dengan streak kuadran pada koloni yang membentuk zona ungu, lalu isolat kapang kitinolitik yang diperoleh diuji daya kitinolitiknya dengan metode difusi sumur. Media yang digunakan adalah acidified potato dextrose agar dengan penambahan MgSO4 0,5%, Na2HPO4 0,5%, kitin koloidal 0,5% dan indikator bromocresol purple 0,01%. Pada hasil pemurnian diperoleh dua isolat kapang. Uji daya dilakukan pada tiap isolat dan diperoleh satu isolat kapang yang menunjukkan daya kitinolitik yang paling kuat dengan ciri hifa tidak bersekat, zygospora, koloni berwarna abu-abu dan serupa kapas, serta konidiofor yang bercabang. Selanjutnya, isolat tersebut diidentifikasi dengan metode Polymerase Chain Reaction (PCR). Hasil identifikasi isolat adalah Mucor circinelloides, kapang ini dapat ditemukan pada udara dan lingkungan. / Black tiger shrimp (Penaeus monodon) shell waste contains chitin which is insoluble in water because it’s bound with protein and amino acid. However, chitin has many applications such as in foods, agricultural, pharmaceuticals, and buildings, so it should be converted to its derivative to be used optimally. The objectives in this study are isolating and identifying chitinolityc mold which shows the strongest chitinolytic activity isolated from black tiger shrimp shell waste. The shrimp shell powder was obtained by sun drying, and size reduction, and then it was rotted. Pour plate method of the rotten shrimp shell powder on the level of 10-3, 10-4, and 10-5 was done, later it was isolated and purified by streak quadran method. The media that had been used was acidified potato dextrose agar with the addition of MgSO4 0.5%, Na2HPO4 0.5%, colloidal chitin 0.5%, and bromocresol purple 0.01%. Two molds isolate was obtained and the test of chitinolytic activity was run against those isolates. The strogest mold isolate characteristics are the hyphae are not insulated, zygospore, grey colony and cotton-like colony, and also irregular branch of conodiofor. Furthermore, that isolate was identified by Polymerase Chain Reaction (PCR) method. Result of the PCR method showed that the isolate is Mucor circinelloides, which was found on the air and environment in the previous study.
