Abstract
Tanaman petai (Parkia speciosa Hassk.) merupakan tanaman yang dapat tumbuh di Asia Tenggara seperti Indonesia. Tanaman petai dapat dimanfaatkan sebagai obat tradisional, yaitu sebagai agen antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri crude extract etanol 96% daun petai terhadap bakteri Streptococcus pyogenes, Staphylococcus aureus, dan Escherichia coli. Metode ekstraksi yang digunakan yaitu maserasi dengan menggunakan pelarut etanol 96%, serta metode pengujian aktivitas antibakteri menggunakan metode well diffusion. Penelitian ini menggunakan tiga konsentrasi crude extract daun petai yaitu 200.000 ppm, 400.000 ppm, 800.000 ppm, kontrol positif (injeksi gentamisin), dan kontrol negatif (DMSO). Hasil uji skrining fitokimia crude extract daun petai mengandung metabolit sekunder berupa alkaloid, tanin, saponin, dan steroid. Pengujian pada bakteri S. pyogenes dengan konsentrasi 200.000 ppm dan 400.000 ppm serta konsentrasi 400.000 ppm dan 800.000 ppm tidak ada perbedaan yang signifikan karena terdapat angka yang tumpang tindih. Sedangkan pengujian pada bakteri S. aureus pada konsentrasi 200.000 ppm dan 400.000 ppm tidak ada perbedaan yang signifikan karena terdapat angka yang tumpang tindih. / Petai (Parkia speciosa Hassk.) is a plant that can grow in Southeast Asia such as Indonesia. Petai plants can be used as traditional medicine, namely as an antibacterial agent. This study aims to determine the antibacterial activity of 96% ethanol crude extract of petai leaves against Streptococcus pyogenes, Staphylococcus aureus, and Escherichia coli bacteria. The extraction method used is maceration using 96% ethanol solvent, and the antibacterial activity testing method using the well diffusion method. This study used three concentrations of petai leaf crude extract, namely 200,000 ppm, 400,000 ppm, 800,000 ppm, positive control (gentamicin injection), and negative control (DMSO). The results of the phytochemical screening test of crude petai leaf extract contain secondary metabolites in the form of alkaloids, tannins, saponins, and steroids. Tests on S. pyogenes bacteria with concentrations of 200,000 ppm and 400,000 ppm and concentrations of 400,000 ppm and 800,000 ppm there were no significant differences because there were overlapping figures. While testing on S. aureus bacteria at concentrations of 200,000 ppm and 400,000 ppm there was no significant difference because there were overlapping numbers.
