Abstract

Radikal bebas dalam tubuh manusia dapat dibantu dinetralisir oleh antioksidan alami yang berasal dari tumbuhan. Salah satu tumbuhan yang diduga berpotensi sebagai antioksidan, yaitu daun pepaya jepang (C. aconitifolius). Penelitian ini bertujuan untuk mengetahui kandungan senyawa metabolit sekunder dan aktivitas antioksidan ekstrak etanol 96% daun pepaya jepang (C. aconitifolius) dengan metode DPPH yang dinyatakan dalam parameter IC50 (Inhibitory Concentration) dan AAI (Antioxidant Activity Index). Simpilisia daun pepaya jepang diekstraksi menggunakan pelarut etanol 96% dengan metode maserasi, kemudian pada ekstrak dilakukan penapisan fitokimia, dan selanjutnya dilakukan uji aktivitas antioksidan dengan menggunakan metode DPPH. Dari proses ekstrasi diperoleh rendemen sebesar 16,1834%. Hasil dari penapisan fitokimia ekstrak mengandung senyawa flavonoid, tanin, saponin, fenol, alkaloid, dan steroid. Hasil uji aktivitas antioksidan menunjukkan bahwa ekstrak etanol 96% daun pepaya jepang (C. aconitifolius) memiliki nilai IC50 sebesar 145,3855 μg/mL (lemah) dan nilai AAI sebesar 0,4127 (lemah). Berdasarkan hasil yang diperoleh, ekstrak etanol 96% daun pepaya jepang (C. aconitifolius) memiliki aktivitas antioksidan dengan kategori lemah berdasarkan parameter IC50 dan AAI. / Free radicals in the human body can be assisted neutralized by the natural antioxidants of plant origin. One of the plants that have the potential as antioxidants, namely papaya leaves japan (C. aconitifolius). This study aims to determine the content of secondary metabolite compounds and antioxidant activity of ethanol extract 96% of papaya leaves japan (C. aconitifolius) by the method of DPPH scavenging activity, which is expressed in the parameters of the IC50 (Inhibitory Concentration) and AAI (Antioxidant Activity Index). Simpilisia papaya leaves japan extracted using ethanol 96% solvent by the method of maceration, then the extract is carried out phytochemical screening, and further test the antioxidant activity by using DPPH. The process of extraction obtained yield of 16,1834%. The results of phytochemical screening of the extract containing flavonoids, tannins, saponins, phenols, alkaloids, and steroids. The results of the antioxidant activity test showed that the ethanol extract 96% of papaya leaves japan (C. aconitifolius) have IC50 values of 145,3855 µg/mL (weak) and the value of AAI by 0,4127 (weak). Based on the results obtained, the extract of ethanol 96% of papaya leaves japan (C. aconitifolius) has antioxidant activity with the category of weak within the parameters of the IC50and AAI.
