Abstract

Prebiotik adalah substrat yang dapat mendukung pertumbuhan bakteri probiotik seperti Lactobacillus sp. dan Bifidobacterium sp.. Penelitian ini bertujuan untuk melihat pertumbuhan dan aktivitas fermentasi berbagai prebiotik terhadap SU-KC1a dan STBM; uji pertumbuhan SU-KC1a dan L. plantarum F75 sebagai pembanding pada media MRS+mupirocin; uji poliformisme pada SU-KC1a pada kondisi mikroaerofilik dan anaerobik obligat. Metode penelitian terdiri dari (1) uji pertumbuhan L. plantarum F75 pada media MRS+mupirocin dalam kondisi anaerobik obligat dan mikroaerofilik di suhu 37oC selama 72 jam, (2) pertumbuhan SU-KC1a diinkubasi dalam kondisi mikroaerofilik dan anaerobik obligat dengan media MRS+mupirocin selama 72 jam di suhu 37oC, (3) uji aktivitas fermentasi SU-KC1a dan STBM yang ditumbuhkan pada media MRS (-glukosa)+prebiotik dan kontrol positif dengan penambahan masing-masing 2% FOS, GOS, inulin, maltodextrin, glukosa, fruktosa, laktosa dengan menggunakan indikator fermentasi bromocresol purple selama 72 jam di suhu 37oC secara anaerobik obligat, (4) uji enumerasi pertumbuhan SU-KC1a dan STBM pada media MRS (2% prebiotik) dan media MRS-kontrol positif selama 72 jam di suhu 37oC secara anaerobik obligat. Dari hasil penelitian tidak didapat adanya pertumbuhan L. plantarum F75 pada media MRS+mupirocin; isolat SU-KC1a mengalami perubahan morfologi dari rod-shaped pendek menjadi panjang pada kondisi masing-masing anaerobik obligat dan mikroaerofilik; isolat SU-KC1a dan STBM memiliki aktivitas fermentasi terhadap semua media MRS (-glukosa)+prebiotik dan MRS-kontrol positif yang diindikasikan berdasarkan perubahan warna dan pH; hasil enumerasi penambahan 2% prebiotik pada pertumbuhan SUK-C1a didapat GOS> Inulin> FOS> maltodextrin> laktosa> glukosa> fruktosa; pengamatan ukuran koloni didapat lebih besar pada oligosakarida daripada di,- monosakarida. Kesimpulan dari penelitian ini adalah: SU-KC1a sebagai spesies L. plantarum memiliki resistensi terhadap mupirocin, bersifat polimorfik dalam kondisi pertumbuhan yang berbeda, SU-KC1a dan STBM dapat tumbuh paling baik pada prebiotik GOS, memiliki aktivitas fermentasi, dan kecepatan fermentasi lebih cepat pada STBM daripada SU-KC1a./Prebiotics is a substrates that could support growth of probiotic bacteria such as Lactobacillus sp. and Bifidobacterium sp.. This research aims to observe growth and fermentation activity of SU-KC1a and STBM in a various prebiotics; growth test of SU-KC1a and L. plantarum F75 as comparison on MRS+mupirocin media; polyformism test of SU-KC1a under microaerophilic and obligate anaerobic conditions. This research method consisted of (1) growth test of L. plantarum F75 on MRS+mupirocin media under obligate anaerobic and microaerophilic conditions at 37oC for 72 hours, (2) SU-KC1a is incubated under microaerophilic and obligate anaerobic conditions with MRS+mupirocin medium. for 72 hours at 37oC, (3) fermentation activity test of SU-KC1a and STBM was grown on MRS (-glucose)+ prebiotic media and positive control with the addition of 2% FOS, GOS, inulin, maltodextrin, glucose, fructose, lactose respectively using bromocresol purple fermentation indicator for 72 hours at 37oC obligate anaerobically, (4) growth enumeration test of SU-KC1a and STBM on MRS medium (2% prebiotic) and positive control MRS medium for 72 hours at 37oC in obligate anaerobes condition. The result shows that, there was no growth of L. plantarum F75 on MRS+mupirocin media; SU-KC1a underwent morphological changes from short rod-shaped to long under conditions of obligate anaerobic and microaerophilic, respectively; SU-KC1a and STBM had fermentative activity against all MRS (-glucose)+prebiotic and MRS-positive control media indicated by color and pH changes; the best growth in enumeration with the addition of 2% prebiotic of SUK-C1a was obtained in GOS> Inulin> FOS> maltodextrin> lactose> glucose> fructose; Colony size observation was found to be larger in oligosaccharides than in monosaccharides. The conclusions of this study are: SU-KC1a as a species of L. plantarum has resistance to mupirocin, is polymorphic in different growing conditions, SU-KC1a and STBM can grow best on prebiotic GOS, have fermentation activity, and fermentation speed is faster in STBM than SU-KC1a.
