Abstract
Lactobacillus casei merupakan salah satu bakteri yang berpotensi sebagai probiotik dengan aktivitas antibakteri. Pada penelitian ini, bakteri dari stok kultur Lactobacillus casei DRZ97/L3 diidentifikasi secara morfologi dan molekuler. Selanjutnya, dilakukan evaluasi akivitas antibakteri dari cell free supernatant (CFS)
L. casei DRZ97/L3 terhadap pertumbuhan E. coli dan S. aureus. Pengujian meliputi pengaruh suhu dan pH terhadap aktivitas antibakteri dari CFS. Identifikasi morfologi dilakukan berdasarkan pewarnaan gram, endospora dan tahan asam. Selain itu, dilakukan pula uji katalase. Proses identifikasi molekular yang dilakukan meliputi ekstraksi DNA, amplifikasi gen 16S rRNA dengan primer universal, sekuensing dan analsis BLAST. Hasil penelitian menunjukkan bahwa aktivitas antibakteri L. casei DRZ97/L3 terhadap pertumbuhan bakteri E. coli dan S. aureus dipengaruhi oleh pH, tetapi tidak oleh suhu. Aktivitas penghambatan terbaik didapatkan pada keadaan pH sangat rendah, yaitu pH 3. Suhu tidak mempengaruhi aktivitas antibakteri karena komponen antibakteri diduga tahan terhadap panas. Hasil analisis 16S rRNA menunjukan L. casei DRZ97/L3 memiliki kekerabatan yang tinggi dengan L. paracasei strain L1 dan L. casei strain YZU01. / Lactobacillus casei is one of the bacteria that has the potential as a probiotic. One of the characteristics of probiotics is that they have antibacterial activity. In this study, an evaluation of the inhibition (antibacterial) of Lactobacillus casei DRZ97/L3 was carried out on the growth of E. coli and S. aureus. The antibacterial activity of L. casei DRZ97/L3 was evaluated by the well diffusion method; time- kill test (co-culture); Broth macrodilution assay (BMA); and 16s rRNA molecular assay. Molecular tests were carried out by DNA extraction, PCR amplification with universal primers. The results of the well-diffusion method showed that the antibacterial activity of L. casei DRZ97/L3 showed positive results in inhibiting the growth of E. coli and S. aureus bacteria under low pH conditions. The best results are at very low pH conditions, namely at pH 3. The results of the 16S analysis show the species in the form of L. casei or L. paracasei. Temporary conclusion is that L. casei DRZ97/L3 has potential as a probiotic
