Abstract

Lactiplantibacillus plantarum (Lpb. plantarum) merupakan bakteri asam laktat yang umum digunakan sebagai probiotik.Dalam aplikasi penggunaan probiotik secara industri, terdapat kondisi dan faktor yang dapat menyebabkan stres sehingga memengaruhi viabilitas probiotik. Penelitian ini memiliki dua komponen: (1) Identifikasi secara bioinformatika gen-gen terkait respon terhadap stres pada Lpb. plantarum strain SU-KC1a yang telah diisolasi dari ASI dengan menggunakan anotasi Whole Genome Sequencing (WGS). Secara lebih lanjut, gen terkait respon terhadap stres oksidatif dianalisis dan dibandingkan dengan tiga strain Lpb. plantarum lain. (2) Percobaan ketahanan SU-KC1a terhadap paparan H2O2. Metode yang digunakan adalah: (1) Perakitan data WGS dengan menggunakan software Flye dan Medaka. Proses anotasi dilakukan menggunakan software Prokka. (2) Percobaan ketahanan stres oksidatif SU-KC1a dengan penambahan 0,5% H2O2 ke dalam kultur pada fase stasioner dan eksponensial (pada kondisi aerobik, anaerobik, dan mikroaerofilik) dengan dan tanpa penambahan mangan dalam medium untuk dienumerasi. Hasil yang diperoleh adalah terdapat gen-gen terkait respon terhadap stres pada SU-KC1a. Gen yang teranotasi berkaitan dengan respon terhadap stres secara umum, stres suhu rendah dan suhu tinggi, stres pH asam, stres tekanan osmotik, stres oksidatif, dan protein dengan fungsi chaperone dan protease. Gen-gen tersebut juga ditemukan pada Lpb. plantarum lainnya. Berdasarkan analisis yang dilakukan, diperkirakan bahwa gen terkait stres oksidatif terkonservasi pada spesies Lpb. plantarum. Selain itu, pada percobaan ketahanan stres oksidatif SU-KC1a diperkirakan bahwa pada fase stasioner dalam kondisi pertumbuhan mikroaerofilik dengan penambahan mangan memiliki pertumbuhan terbaik setelah inkubasi dengan H2O2 selama 40 menit. Sementara itu, pertumbuhan tidak teramati pada kultur dalam fase eksponensial setelah 30 menit inkubasi dengan H2O2. / Lactiplantibacillus plantarum (Lpb. plantarum) is a lactic acid bacterium commonly used in functional food industry as probiotics. In industrial processes, there are stress factors that could affect probiotics’ viability. To address that, this research was done in two parts: (1) Identification of genes relating to stress response in Lpb. plantarum strain SU-KC1a isolated from human breast milk using Whole Genome Sequencing (WGS) annotation. Furthermore, genes associated with oxidative stress response were analyzed and compared to three other Lpb. plantarum strains. (2) Examination of SU-KC1a resistance towards H2O2 stress. The methods done in this research were: (1) Assembly of WGS using Flye and Medaka softwares. Prokka software was also used to annotate SU-KC1a WGS. (2) Examination of SU-KC1a resistance towards H2O2 stress was done by adding 0,5% H2O2 into culture in stationary and exponential phase (incubated in aerobic, anaerobic, and microaerophilic conditions) with and without manganese in medium for enumeration. Genes related to stress response were identified in SU-KC1a. The annotated genes are associated with responses for universal stress, heat and cold stress, acid stress, osmotic stress, oxidative stress, and also chaperone and protease protein activated in stress condition. It could be inferred that genes relating to oxidative stress response are conserved in Lpb. plantarum species. In the evaluation of SU-KC1a resistance towards H2O2 stress, it was found that in stationary phase, SU-KC1a culture grown in microaerophilic condition with added manganese in medium presumably had the best growth after incubation with H2O2 for 40 minutes. Meanwhile, growth was not evident in exponentially grown SU-KC1a culture after 30 minutes of incubation period with H2O2.
