Abstract

The textile industry in Indonesia has a high contribution to the country's economic growth, not only for the domestic market but also for foreign markets. However, one thing that needs to be considered in the textile industry is the waste that it produces. Textile dyes disposed into water bodies have the potential to be carcinogenic and mutagenic agents. Biological decolorization process can be applied by utilizing various indigenous microorganisms, especially from fermented foods such as tapai, because they are considered safer. This study aims to examine the potential of yeast decolorization isolated of tapai yeast against textile dyes, analyze the optimal conditions needed, analyze decolorization metabolites, and identify the yeast. From six fungi isolated from tapai, it was discovered that isolate 2 had the highest decolorization ability (97,94%) at the dye concentration of 500 ppm and was still able to perform the decolorization process well at increased dye concentration of 4000 ppm (93,11%) within 120 hours. From the analysis of the decolorization mechanism, it was found that isolate 2 decolorized textile dyes by bioaccumulation and biosorption, but the biodegradation activity was very low. Through optimization with the RSM method, the optimum conditions needed by isolate 2 to decolorize textile dyes were obtained at 30,30 oC, pH 7, and with inoculum volume of 3,07% (v/v) for 74,4 hours. The results of the analysis performed using the TLC method and UVVis spectrophotometer showed no formation of new metabolites and confirmed the assumption that the main decolorization process of isolates was bioaccumulation. From the results of morphological identification and biochemical activity tests, it was suspected that isolate 2 was yeast Hyphopichia burtonii./ Industri tekstil di Indonesia memiliki kontribusi tinggi terhadap pertumbuhan ekonomi negara, tidak hanya bagi pasar dalam negeri namun juga bagi pasar luar negeri. Namun, salah satu hal yang perlu diperhatikan pada industri tekstil adalah limbah yang dihasilkannya. Limbah pewarna teksil yang dibuang ke perairan berpotensi sebagai agen karsinogenik dan mutagenik. Proses dekolorisasi secara biologis dapat diterapkan dengan memanfaatkan berbagai mikroorganisme indigenous, khususnya yang berasal dari makanan fermentasi seperti tapai, karena dianggap lebih aman. Penelitian ini ditujukan untuk menguji potensi dekolorisasi khamir yang diisolasi dari ragi tapai terhadap pewarna tekstil, mengetahui kondisi optimal yang dibutuhkan, menganalisis metabolit hasil dekolorisasi, dan mengidentifikasi khamir tersebut. Dari enam isolat fungi, isolat 2 diketahui memiliki kemampuan dekolorisasi tertinggi (97,94 %) pada konsentrasi pewarna 500 ppm dan masih mampu melakukan proses dekolorisasi dengan baik pada konsentrasi pewarna yang ditingkatkan menjadi 4000 ppm (93,11 %) dalam waktu 120 jam. Dari hasil analisis mekanisme dekolorisasi, diketahui bahwa isolat 2 mendekolorisasi pewarna tekstil dengan cara bioakumulasi dan biosorpsi, namun aktivitas biodegradasinya sangat rendah. Melalui optimasi dengan metode RSM, diperoleh kondisi optimal yang dibutuhkan oleh isolat 2 untuk mendekolorisasi pewarna tekstil yaitu pada suhu 30,30 oC, pH 7, dan volume inokulum 3,07 % (v/v) selama 74,4 jam. Hasil analisis dengan metode TLC maupun spektrofotometer UV-Vis menunjukkan tidak terbentuknya metabolit baru dan mengkonfirmasi dugaan bahwa proses dekolorisasi utama isolat yaitu dengan bioakumulasi. Dari hasil identifikasi morfologi dan uji aktivitas biokimia, diduga isolat 2 merupakan khamir Hyphopichia burtonii.
