Abstract
Radikal bebas merupakan suatu molekul yang mengandung satu atau lebih dari satu elektron yang tidak saling berpasangan. Pada saat didalam tubuh, radikal bebas menjadi senyawa yang reaktif yang dapat mengakibatkan terjadinya penyakit. Antioksidan dapat menghambat terjadinya pembentukan radikal bebas didalam tubuh dengan menjadi pendonor elektron untuk radikal bebas. Salah satu tumbuhan yang diduga berpotensi sebagai antioksidan yaitu kulit batang biwa (Eriobotrya japonica) (Thunb.) Lindl.). Penelitian ini bertujuan untuk mengetahui kandungan senyawa metabolit sekunder dan aktivitas antioksidan ekstrak etanol 96% kulit batang biwa (Eriobotrya japonica) (Thunb.) Lindl.) dengan metode DPPH yang dinyatakan dalam parameter IC50 (Inhibition concentration 50%). Simplisia kulit batang biwa di ekstrak menggunakan pelarut 96% dengan metode maserasi dan dilakukan skrining fitokimia dan dilanjutkan dengan uji aktivitas antioksidan menggunakan metode DPPH. Dari proses ekstraksi diperoleh rendamen sebesar 12,295%. Hasil dari skrining fitokimia ekstrak mengandung senyawa alkaloid, flavonoid, saponin, tanin, fenol dan terpenoid. Hasil uji aktivitas antioksidan secara kuantitatif menunjukkan bahwa ektrak etanol 96% kulit batang biwa (Eryobotrya japonica) (Thunb.) Lindl.) memiliki nilai IC50 sebesar 42,87 μg/mL. Berdasarkan hasil yang diperoleh menggunakan parameter IC50 ekstrak etanol 96% kulit batang biwa (Eriobotrya japonica) (Thunb.) Lindl.) memiliki aktivitas antioksidan dengan kategori kuat. / Free radicals are molecules that contain one or more unpaired electrons. In the body, free radicals become reactive compounds that can cause disease. Antioxidants can inhibit the formation of free radicals in the body by becoming electron donors for free radicals. One of the plants that is thought to have potential as an antioxidant is the bark of the loquat (Eriobotrya japonica) (Thunb.) Lindl.). This study aims to determine the content of secondary metabolites and antioxidant activity of 96% ethanol extract of biwa (Eriobotrya japonica) (Thunb.) Lindl.) stem bark using the DPPH method which is stated in the IC50 parameter (50% Inhibition Concentration). Biwa stem bark simplicia was extracted using 96% solvent by maceration method and phytochemical screening was carried out and continued with antioxidant activity test using the DPPH method. From the extraction process, the yield of 12.295% was obtained. The results of phytochemical screening of extracts contained alkaloids, flavonoids, saponins, tannins, phenols and terpenoids. The results of the quantitative antioxidant activity test showed that the 96% ethanol extract of the bark of the loquat (Eryobotrya japonica) (Thunb.) Lindl.) had an IC50 value of 42.87 μg/mL. Based on the results obtained using the IC50 parameter, 96% ethanol extract of the bark of the loquat (Eriobotrya japonica) has antioxidant activity with a strong category.
