Abstract
Daun tekelan (Chromolaena odorata (L) R.M.King & H.Rob.) merupakan salah satu
tanaman yang memiliki khasiat obat. Daun tekelan memiliki senyawa metabolit
yang berpotensi sebagai antibakteri terhadap bakteri Staphylococcus aureus. Oleh
karena itu, tujuan penelitian ini adalah untuk membuat sediaan face toner dengan
bahan aktif ekstrak etanol 96% daun tekelan yang memenuhi syarat mutu fisik serta
memiliki daya aktivitas antibakteri. Ekstrak daun tekelan dibuat dengan metode
maserasi dengan pelarut etanol 96% dan uji aktivitas antibakteri menggunakan
metode sumuran agar yang diinkubasi pada suhu 37℃. Selanjutnya dibuat sediaan
face toner dengan variasi konsentrasi ekstrak 1,25%, 2,5% dan 5%. Dilakukan
evaluasi terhadap sediaan face toner yang dibuat, meliputi uji organoleptis, uji pH,
uji homogenitas, uji viskositas dan uji bobot jenis. Sediaan kemudian disimpan
selama 12 hari pada suhu ruang. Hasil ekstraksi diperoleh rendemen sebesar 25,40%
dan memiliki kandungan senyawa metabolit sekunder yaitu, alkaloid, fenol,
flavonoid, tanin, saponin dan steroid. Hasil pembuatan sediaan face toner
menghasilkan sediaan yang memenuhi persyaratan uji mutu fisik yang baik. Dari
hasil uji stabilitas 12 hari, terlihat adanya ketidakstabilan setelah penyimpanan
dimana terlihat adanya ketidakstabilan perubahan warna dan aroma pada formulasi
kedua dan ketiga dengan konsentrasi ekstrak 2,5% dan 5%. Hal tersebut
menandakan ketidakstabilan sediaan saat penyimpanan. Pengujian aktivitas
antibakteri ekstrak 1,25% setara dengan 22,96 ppm, ekstrak 2,5% setara dengan
27,41 ppm dan ekstrak 5% setara dengan 64,44 ppm antibiotik gentamisin.
Pengujian aktivitas antibakteri sediaan toner menunjukan bahwa sediaan memiliki
aktivitas antibakteri terhadap bakteri Staphylococcus aureus. Dimana, pada formula
1 setara dengan 22,96 ppm antibiotik gentamisin, formula 2 setara dengan 27,41
ppm serta formula 3 setara dengan 64,44 ppm antibiotik gentamisin. Berdasarkan
hasil yang diperoleh, dapat disimpulkan bahwa ekstrak etanol 96% daun tekelan
dapat diformulasikan menjadi sediaan toner dan memiliki aktivitas antibakteri
terhadap bakteri Staphylococcus aureus./ Tekelan leaf (Chromolaena odorata (L) R.M.King & H.Rob.) is one of the plants
that have medicinal properties. Tekelan leaves have metabolite compounds that
have the potential to be antibacterial against Staphylococcus aureus bacteria.
Therefore, this study aims to make a face toner preparation with the active
ingredient ethanol extract of 96% tackle leaves that meet the physical qualification
requirements and have antibacterial activity. The tackle leaf extract was made by
maceration method with 96% ethanol solvent and antibacterial activity test using
the incubated agar well method at 37 °C. Furthermore, face toner preparations are
made with variations in extract concentrations of 1.25%, 2.5%, and 5%. Evaluation
of the face toner preparations is carried out, including organoleptic tests, pH tests,
homogeneity tests, viscosity tests, and type weight tests. The preparations were then
stored for 12 days at room temperature. The extraction results obtained a yield of
25.40% and contain secondary metabolites, namely, alkaloids, phenols, flavonoids,
tannins, saponins, and steroids. The results of making face toner preparations
produce preparations that meet the requirements of a good physical quality test.
From the results of the 12-day stability test, it was seen that there was instability
after storage where there was an unstable change in color and aroma in the second
and third formulations with extract concentrations of 2.5% and 5%. This indicates
the instability of the preparation during storage. The antibacterial activity test of
1.25% extract was equivalent to 22.96 ppm, 2.5% extract was equivalent to 27.41
ppm and 5% extract was equivalent to 64.44 ppm of the antibiotic gentamicin.
Testing the antibacterial activity of toner preparations showed that the
preparations had antibacterial activity against Staphylococcus aureus bacteria.
Where formula 1 is equivalent to 22.96 ppm of gentamicin antibiotics, formula 2 is
equivalent to 27.41 ppm and formula 3 is equivalent to 64.44 ppm of gentamicin
antibiotics. Based on the results obtained, it can be concluded that the 96% ethanol
extract of tekelan leaves can be formulated into toner preparations and has
antibacterial activity against Staphylococcus aureus bacteria.
