Abstract
Dunia kesehatan saat ini banyak membahas mengenai radikal bebas (free radical) dan antioksidan. Radikal bebas merupakan suatu molekul yang memiliki satu atau lebih elektron tidak berpasangan. Radikal bebas memiliki sifat yang reaktif sehingga dapat bereaksi dengan berbagai molekul lain seperti lipid, protein dan DNA. Antioksidan merupakan senyawa yang dapat menghambat radikal bebas, dengan cara mengikat radikal bebas dan molekul yang sangat reaktif sehingga kerusakan sel dapat dihambat. Salah satu tumbuhan yang diduga memiliki potensi aktivitas antioksidan adalah daun C. argentea. Penelitian ini bertujuan untuk mengetahui kandungan senyawa metabolit sekunder, total fenolik, total flavonoid dan aktivitas antioksidan ekstrak etil asetat daun C. argentea. Penentuan kuantitatif total fenol dinyatakan sebagai Gallic Acid Equivalent (GAE) mg/gram ekstrak, kadar total flavonoid dinyatakan sebagai Quercetin Equivalen (QE), dan aktivitas antioksidan dengan metode DPPH (2, 2-Diphenyl-1-picrylhydrazyl) yang dinyatakan dalam istilah IC50 (Inhibition Concentration). Hasil dari penelitian ini adalah rendamen dari ekstraksi diperoleh sebesar 5,0806% b/b. Hasil penapisan fitokimia menunjukkan adanya kandungan senyawa flavonoid, alkaloid, fenolik, tanin, dan steroid dalam ekstrak. Selain itu didapatkan juga total fenolik sebesar 807,324 mg GAE/g ekstrak, sedangkan kandungan total flavonoid adalah 86,35 mg QE/g ekstrak. Nilai IC50 yang diperoleh dari hasil pengujian aktivitas antioksidan ekstrak etil asetat daun C. argentea adalah 34,9661 ppm. Berdasarkan hasil yang diperoleh maka ekstrak etil asetat daun C. argentea memiliki aktivitas antioksidan yang kuat. / The health science is currently discussing about free radicals and antioxidants. Free radicals are molecules that have one or more unpaired electrons. Free radicals have reactive properties so they can react with various other molecules such as lipids, proteins and DNA. Antioxidants are compounds that can inhibit free radicals by binding to free radicals and highly reactive molecules so that cell damage can be inhibited. One of the plants with is thought to have potential antioxidant activity is C. argentea leaves. This study aims to determine the content of secondary metabolites, total phenolic compounds, total flavonoids and antioxidant activity of the ethyl acetate extract of C. argentea leaves. Meanwhile quantitative determination of total phenolic content was expressed as Gallic Acid Equivalent (GAE) mg/gram extract, total flavonoid content was expressed as Quercetin Equivalent (QE), and antioxidant activity by DPPH method (2, 2-Diphenyl-1-picrylhydrazyl) expressed in terms of IC50 (Inhibition Concentration). The result of this research is that the yield of the extraction is 5.0806% w/w. The results of phytochemical screening showed the presence of flavonoid, alkaloid, phenolic, tannin, and steroid compounds in the extract. In addition, the total phenolic content was 807,324 mg GAE/g extract, while the total flavonoid content was 86,35 mg QE/g extract. The IC50 value obtained from the results of testing the antioxidant activity of the ethyl acetate extract of C. argentea leaves was 34.9661 ppm. Based on the results obtained, the ethyl acetate extract of C. argentea leaves has strong antioxidant activity.
