Abstract
The aquatic animals such as crustaceans (mud crab) are known as a major source of carotenoids. Few studies have been carried out that asthaxanthin and its esters were found to be the major carotenoids in crustaceans. The objective of this research were to determine the best maceration temperature from each solvent that used to extract carotenoid from mud crab shell by determining of the highest yield and total carotenoid content, and to study the potency of carotenoid extract as antioxidant, antibacterial, and triterpenoid content. In this research, carotenoid was extracted by using three different temperatures (30'C, 45'C, and 60'C) and three different solvents (hexane, isopropanol, and solvent mixture of hexane and isopropanol 1 : 1). Total carotenoid content from extract were determined by using formula from Sacchindra et al. (2006), the activity of antioxidant was determined by using DPPH method, the activity of antimicrobial was determined by using well-diffusion method, and triterpenoid content was determined by using Liebermann-Burchard method. Result showed that 45'C was the best maceration temperature and the best solvent to extract the carotenoid from mud crab is hexane. The IC50 for hexane w as 11471.5 ppm, the IC50 for isopropanol was 15293 ppm, and IC50 for mixture solvent (hexane ; isopropanol) was 13041 ppm. Three types of carotenoid extract contained triterpenoid, however the did not have an antibacterial activity towards both Gram - Gram+.
