Abstract
Gout merupakan penyakit yang disebabkan oleh tingginya kadar asam urat di dalam darah (hiperurisemia). Dalam produksi asam urat, enzim xantin oksidase berperan sebagai katalisator reaksi oksidasi hipoxantin menjadi xantin dan membentuk asam urat. Untuk mencegah dan mengobati penyakit ini, diperlukan inhibitor untuk menghambat aktivitas xantin oksidase. Xantin oksidase dapat dihambat dengan menggunakan metabolit sekunder yang ditemukan pada tanaman. Ficus callosa Warb merupakan tanaman yang memiliki metabolit sekunder flavonoid. Tujuan penelitian ini adalah untuk mengetahui potensi ekstrak daun Ficus callosa Warb sebagai inhibitor xantin oksidase yang dinyatakan dalam nilai persentase penghambatan dan kemampuan ekstrak Ficus callosa Warb menghambat pembentukan asam urat yang dinyatakan dalam nilai IC50. Ekstraksi dilakukan dengan metode refluks dengan pelarut bertingkat n-heksana, etil asetat, dan etanol 96%, ekstrak yang memenuhi standar kemudian dilakukan skrining fitokimia dan uji inhibisi xantin oksidase. Berdasarkan penelitian yang telah dilakukan, ekstrak daun Ficus callosa Warb memiliki metabolit sekunder flavonoid yang dapat digunakan sebagai inhibitor alami enzim xantin oksidase. Ekstrak etanol 96% daun Ficus callosa Warb memiliki kemampuan penghambatan paling baik pada konsentrasi 125 ppm. / Gout is a disease caused by high levels of uric acid in the blood (hyperuricemia). In the production of uric acid, the enzyme xanthine oxidase acts as a catalyst for oxidation reaction of hypoxanthine to xanthine and forms uric acid. To prevent and treat this disease, inhibitors are needed to inhibit xanthine oxidase activity. Xanthine oxidase can be inhibited by using secondary metabolites found in plants. Ficus callosa Warb is a plant that has secondary metabolites of flavonoids. The purpose of this study was to determine the potential of Ficus callosa Warb leaf extract as a xanthine oxidase inhibitor expressed in inhibition percentage values and the ability of Ficus callosa Warb extract to inhibit uric acid formation expressed in IC50 values. Extraction was carried out by reflux method with solvent n-hexane, ethyl acetate, and 96% ethanol, extracts that meet the standards were then subjected to phytochemical screening and xanthine oxidase inhibition tests. Based on research that has been done, Ficus callosa Warb leaf extract has secondary metabolites of flavonoids which can be used as natural inhibitors of the xanthine oxidase enzyme. The 96% ethanol extract of Ficus callosa Warb leaves had the best inhibitory ability at a concentration of 125 ppm.
