Abstract

Radikal bebas merupakan senyawa reaktif yang memiliki satu atau lebih elektron yang tidak berpasangan. Dalam kehidupan, paparan radikal bebas tidak bisa terhindarkan dari manusia, sehingga perlu senyawa antioksidan untuk menangkal radikal bebas. Dampak yang dapat terjadi jika radikal bebas meningkat antara lain seperti stres oksidatif yang memicu terjadinya penyakit degeneratif. Castanopsis tungurrut merupakan tumbuhan yang diduga memiliki aktivitas antioksidan. Penelitian ini bertujuan untuk mengeksplorasi potensi ekstrak batang C.tungurrut sebagai antioksidan melalui mekanisme penghambatan DPPH dan reduksi kompleks ion besi. Ekstrak batang C.tungurrut dilakukan beberapa pengujian yaitu penapisan fitokimia, kandungan fenol dan flavonoid total, dan aktivitas antioksidan menggunakan metode DPPH dan FRAP. Hasil yang diperoleh dari fenolik ekstrak batang C. tungurrut etanol sebesar 514. 7984 ± 23.5663 mg GAE/g ekstrak dan ekstrak batang C. tungurrut etil asetat sebesar 144.8632 ± 3.9688 mg GAE/g. Total flavonoid ekstrak batang C. tungurrut etanol diperoleh 10.0018 ± 3.59669 mg QE/g dan ekstrak etil asetat 24.0398 ± 0,7428 mg QE/g. Hasil ekstrak batang C.tungurrut baik ekstrak etanol dan etil asetat terbukti berpotensi sebagai antioksidan. Ekstrak etanol lebih berpotensi dibandingkan dengan ekstrak etil asetat sebagai penghambat inhibisi DPPH dan agen antioksidan. Ekstrak batang C.tungurrut etanol dan etil asetat memiliki faktor dominan senyawa yang berbeda sebagai antioksidan./ Free radicals are reactive compounds that have one or more unpaired electrons. In life, exposure to free radicals is unavoidable for humans, so antioxidant compounds are needed to counteract free radicals. The impact that can occur if free radicals increase, among others, is oxidative stress, which triggers degenerative diseases. Castanopsis tungurrut is a plant that is thought to have antioxidant activity. This study aims to explore the potential of C. tungurrut stem extract as an antioxidant through the mechanisms of DPPH inhibition and reduction of iron ion complexes. C. tungurrut stem extract was subjected to several tests, namely phytochemical screening, levels of total phenols and flavonoids, and antioxidant activity using the DPPH and FRAP methods. The results obtained from the phenolic stem extract of C. tungurrut ethanol were 514.7984 23.5663 mg GAE/g extract, and the stem extract of C. tungurrut ethyl acetate was 144.8632 3.9688 mg GAE/g. Total flavonoids of C. tungurrut stem extract ethanol obtained 10.0018 3.59669 mg QE/g and ethyl acetate extract 24.0398 0.7428 mg QE/g. The results of C. tungurrut stem extracts showed that both ethanol and ethyl acetate extracts had potential as antioxidants. The ethanol extract is more potent than the ethyl acetate extract as a DPPH inhibitor and antioxidant agent. The ethanol and ethyl acetate stem extracts of C. tungurrut have dominant factors of different compounds as antioxidants.
