Abstract
Radikal bebas dapat berasal dari dalam tubuh (endogen) yang dihasilkan secara
alami dari metabolisme sel-sel tubuh dan dapat berasal dari luar sistem tubuh
(eksogen) yang dihasilkan dari aktivitas lingkungan seperti polusi dan radiasi.
Akumulasi radikal bebas yang berlebih pada tubuh akan menciptakan kondisi
stres oksidatif yang menybabkan ketidakseimbangan radikal bebas dengan
antioksidan. Fenolik dan flavonoid merupakan dua senyawa yang memiliki
peranan sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui kadar
senyawa fenolik dengan menguji kandungan fenolik total, kadar flavonoid dengan
menguji kandungan flavonoid total dan aktivitas antioksidan menggunakan
metode DPPH (2,2-diphenyl-1-picrylhydrazyl) dari etanol, n -heksana, dan fraksi
etil asetat pisang kepok lokal (Musa x paradisiaca L.) asal Nias. Aktivitas
antioksidan akan dinyatakan dalam nilai Inhibition Concentration (IC50) dan
Antioxidant Activity Index (AAI). Hasil pengujian kandungan flavonoid total
tertinggi terdapat pada fraksi n-heksan dengan nilai 944,16±0,3 mg QE/g sampel
sedangkan kandungan fenolik total tertinggi terdapat pada fraksi etil asetat dengan
nilai 331,37±0,25 mg GAE/g sampel. Dari hasil pengujian aktivitas antioksidan
diperoleh bahwa fraksi etanol 70% memiliki aktivitas antioksidan paling besar
dengan nilai IC50 sebesar 20,64 ppm dan nilai AAI sebesar 2,60./Free radicals can come from within the body (endogenous) which are produced
naturally from the metabolism of body cells and can come from outside the body's
system (exogenous) which are produced from environmental activities such as
pollution and radiation. Excessive accumulation of free radicals in the body will
create conditions of oxidative stress, namely the imbalance between free radicals
and antioxidants. Phenolics and flavonoids are two compounds that can act as
antioxidants. This study aimed to determine the concentration of phenolic
compounds by testing the total phenolic content, the concentration of flavonoids
by testing the total flavonoid content and antioxidant activity using the DPPH
(2,2-diphenyl-1-picrylhydrazyl) method from the ethanol, n-hexane, and ethyl
acetate fractions local kepok banana (Musa x paradisiaca L.) from Nias.
Antioxidant activity will be expressed in the Inhibition Concentration (IC50) and
Antioxidant Activity Index (AAI). From the test results, the highest total flavonoid
content was found in the n-hexane fraction with a value of 944,16 ± 0,3 mg QE/g
sample, while the highest total phenolic content was found in the ethyl acetate
fraction with a value of 331,37 ± 0,25 mg GAE/g sample. From the results of
antioxidant activity testing, it was found that the 70% ethanol fraction had the
greatest antioxidant activity with an IC50 value of 20.64 ppm and an AAI value of
2.60.
